Dastarcus helophoroides is the most effective natural enemy against the large-body longhorn beetles attacking trees. Adversity stresses, such as extreme high and low temperature,oxidation, hunger and other environmental stresses, can significantly affect insect survival,development and fecundity. Dastarcus helophoroides has strong environmental adaptability in nature. Heat shock protein(HSP) genes are speculated to possess a pivotal function in response to environmental stress. To clarify the relationship with HSP70 and sHSP in adults,using RT-qPCR to analysis their expression during developmental stages and environmental stress. Therefore, the main results and conclusions presented in this study are as follows:Three full-length HSP70 genes, D.hHSP69.09、D.Hhsp70.11 and D.hHSP71.88, and three intronless sHSP genes, D.hsHSP11.06, D.hsHSP11.10 and D.hsHSP20.99, were obtained from D. helophoroides. The ORFs of genes were 1980, 1967, 1910, 444, 574 and789 bp, which encoded 626, 636, 655, 102, 102 and 185 amino acids, respectively. The predicted molecular masses were 69.09 kDa, 70.11 kDa, 71.88 kDa, 11.06 kDa, 11.09 KDa and20.99 kDa, and the isoelectric points were 5.57, 6.01, 5.93, 9.52, 8.01 and 6.31.The highest expression level of D.hHSP69.09 was found in the firsth larval stage;D.hHSP70.11 and D.hHSP71.88 in the male; D.hsHSP11.10 and D.hsHSP20.99 in the larval stages; D.hsHSP11.06 in the larvae.The highest expression level of D.hHSP69.09 and D.hHSP70.11 was found in ovary;D.hHSP71.88 was found in fat body; D.hsHSP11.06 appeared in the corpus adiposum;D.hsHSP11.10 and D.hsHSP20.99 appeared in the spermary.The expression level of D.hHSP69.09 expressed higher in female adult. The expression level of D.hHSP70.11 and D.hHSP71.88 expressed higher in male adult. The highest expression level of six HSP genes appered at the temperature of 35℃ or 38℃. There were big link between six HSP genes with treatment time at 41℃The expression level of D.hHSP69.09, D.hsHSP11.10 and D.hsHSP20.99 expressed higher in female adult, and D.hHSP70.11 ' D.hHSP71.88 higher in male adult. The highest expression level of six HSP genes appered at the concentration of 20 m M or 30 m M.The female adults showed higher starvation endurance than males as observed. With the time of starvation stress, the expression level of HSP70 genes was decreased. The expression level of D.hHSP69.09 up-regulated at second day; the expression level of D.hHSP70.11 decreased at 4th day, and then increased to the highest expression at the 8th day; the expression level of D.hHSP71.88 increased mildly with the time of starvation stress, and the expression increased sharply at the the 8th day. The highest expression of D.hsHSP11.06,D.hsHSP11.10 and D.hsHSP20.99 appeared at the time of 2d, 10 d and 6d, respectively.In summary, HSP70 and sHSP genes all can respond to various environmental stresses,and they play important roles in the growth, development, reproduction and anti-high temperature, anti-oxidation, anti-hunger and other processes of Dastarcus helophoroides.
