Influence Of Baogan Jiedu Tang (BJT) On Melano-macrophage Centers In Experimental Hepatosis Eel (Anguilla Anguilla)

Objective In the course of aquaculture, the drug abuse and the environmental pollution have brought serve problem of liver diseases to breed aquatics of eel(Anguilla anguilla). The liver-protective Traditional Chinese Medicine Preparation (TCMP) has been applied to prevent and cure the liver diseases in eel production. To further reveal liver-protective effect and mechanism of those kinds of TCMP on liver diseases, the effect on the number and morphology of melano-macrophage centers (MMCs) of spleen and kidney of Baogan Jiedu Tang (BJT) in eel was characterized. Meanwhile, whether the changes of number and morphology on MMCs is suitable as biomarker in liver protection mechanism about agricultural TCMP or not was studied and to make the biomarker more perfect and more persuasive.Methods Thirty-two healthy specimens of eel were randomly divided into four groups (4 fish per group) were studied, and the experiment was carried out in duplicates. In the whole trial course of 8 days all eels without feed and the aerated tap water contained 0.004 mg/1 Cu2+. BJT which was made up of Yinchen, Dahuang, Longdan, Zhizi, and Gancao. Control fish were kept at aerated tap water for 8 days. Cu-poisoned control fish were exposed to waterborne copper concentrations (0.12mg/l) for 4 days, and were kept at aerated tap water in the following 4 days. Low-dosage-drug-treated fish were disposed in the same way as the Cu-poisoned control fish in the first 4 days, and then were administered by 200 mg/1 water solution of BJT in the following 4 days, however, high-dosage-drug-treated fish were administered by 400 mg/1 water solution of BJT in the following 4 days.At the end, fish were killed. Spleen and kidney was dissected out, rinsed with distilled water. The morphology changes and cytochemistry of MMCs of spleen and kidney in eel were assessed by histology, histochemistry. Moreover, the morphology of MMCs of spleen also observed by transmission electron microscopy. HE-stained sections were counted the number of MMCs. Perls stain sections were analyzed 5 parameters changes [Mean MMCs maximum diameter, Mean MMCs minium diameter, Melanin, Lipofuscin and Haemosiderin of MMCs].Results (1) Both splenic MMCs and kidney MMCs contain melanin, lipofuscinin and haemosiderin in each group’s eel by cytochemical observation, but all kinds of pigments of MMCs are different in each group. (2) Compared with the control fish, the patches of MMCs became denser, the number of MMCs did not change obviously, the size of MMCs decreased remarkable (P<0.05), haemosiderin and melanin decreased sharply (P<0.01) and lipofuscin decreased of MMCs of spleen in Cu-poisoned control fish. Compared with the Cu-poisoned control fish, the patches of MMCs became sparser, the number of MMCs did not change obviously, the size of MMCs increased sharply (P<0.01), lipofuscin increased sharply (P<0.01), haemosiderin increased,and melanin decreased of MMCs of spleen in low-dosage-drug-treated fish. Compared with the Cu-poisoned control fish, the changes of 5 parameters [patches, number, size, lipofuscin, melanin] were not obvious, and haemosiderin decreased sharply (P<0.01) of splenic MMCs in high-dosage-drug-treated fish. (3) Compared with the control fish, the patches of MMCs became sparser, the number of MMCs increased remarkable (P<0.05), the size of MMCs remained the samel level, lipofuscin and haemosiderin decreased sharply (P<0.01), melanin decreased of MMCs of kidney in Cu-poisoned control fish. Compared with the Cu-poisoned control fish, the patches of MMCs became even more sparser, the number of MMCs decreased, the size of MMCs increased, lipofuscin and melanin increased remarkable (P<0.05) and haemosiderin increased of MMCs of kidney in low-dosage-drug-treated fish. Compared with the Cu-poisoned control fish, the patches of MMCs became denser, the changes of other 4 parameters [number, size, lipofuscin, melanin] were not obvious, and haemosiderin increased significantly (P<0.01) of MMCs of kidney in high-dosage-drug-treated fish.Conclusion The size of MMCs incresed significantly, lipofuscin incresed sharply and haemosiderin incresed of MMCs of spleen in the low-dosage TCMP group. Haemosiderin decreased significantly and the other parameters did not change evidently of splenic MMCs in the high-dosage TCMP group. The number of MMCs decreased, the size and haemosiderin of MMCs increased, melanin and lipofuscin incresed remarkablely of MMCs of kidney in the low-dosage TCMP group. Haemosiderin increased significantly and the other parameters did not change evidently of kidney MMCs in the high-dosage TCMP group. As a result, the low-dosage BJT is curative against the liver injury by poison.
