Rosa chinensis Jacq.’Old Blush’and Rosa chinensis Jacq.’Slater’s Crimson China’are the most parentages for modern rose cultivars with characteristic of recurrent flowering. Rapid Propagation of the former two roses and regeneration system of Rosa chinensis Jacq var. spontanea (Rehd. et Wils.) Yî’ et Ku were established for rose gene sequencing and transformation.Stems with auxiliary buds of roses were used as explants. MS+2.5 mg/L 6-BA+0.1 mg/LNAA was chosen as the best initial medium; For Rosa chinensis Jacq. var. spontanea (Rehd. et Wils.) Yî’ et Ku,MS+6.0 mg/L 6-BA+0.03 mg/LNAA was the most efficient proliferation medium; MS +0.25mg/L NAA as the most suitable rooting medium; While for Rosa chinensis Jacq.’Old Blush’, MS+4.0 mg/L 6-BA+0.03 mg/L NAA and 1/2MS +0.25mg/L NAA was the best proliferation medium and rooting medium.MS+6.0mg/L 2,4-D was the best callus induction medium for Rosa chinensis Jacq. var. spontanea (Rehd. et Wils.) Yî’ et Ku, and the induction rate was 79.59%; While MS+1.5mg/L NAA+300mg/L L-proline+2.4g/L Gel was most suitable for Rosa chinensis Jacq.’Old Blush’with a rate of 27.91%; The most efficient one for Rosa chinensis Jacq.’Slater’s Crimson China’was MS+4.0 mg/L 2,4-D and induction rate was 20.00%.For Rosa chinensis Jacq. var. spontanea (Rehd. et Wils.) Yî’ et Ku,the somatic embryogenesis can be obtained on MS+3.0mg/L 2,4-D+2.0mg/LTDZ after 12dculture in dark; MS+2.0mg/L 6-BA+ 0.1mg/L NAA was best for somatic embryogenesis maintenance, the induction rate was 30.00%; Somatic embryogenesis can germinate on 1/2MS+2.0mg/L6-BA+0.1mg/L NAA when the PPFD is 150 mol·m-2·s-1. |
