In order to provide guidance for research and explore the better induction methods of antibacterial peptide from Tenebrio molitor, in this thesis, different instar larvae of T. molitor were induced by using different methods, micro injection, feeding and acupuncturing,with E.coli and Staphylococcus aureus, to procude antibacterial peptide. The real-time fluorescent quantitative PCR assay on Tenecin gene of antibacterial peptide from T. molitor was also studied. The results are as follows:1. Effects of the same induction method on different instar larvae of T. molitor are significant different.The antimicrobial activity of antibacterial peptide of 8th,9th and 10th instar larvae induced with E.coli and Staphylococcus aureus were different, while the antimicrobial activity of 9th instar larvaeās antibacterial peptide was the best in all the induction,2. Effects induced by the different bacteria concentration on the larvae of T. molitor are significant different. In all the induction, the effect induced by one of one hundred bacteria concentration was the best.3. The antimicrobial activity of antibacterial peptide induced by the different induction methods on the larvae of T. molitor are significant different. Micro injection was the best method in all the induction.4 The real-time fluorescent quantitative PCR assay on Tenecin gene and Coleoptericin C gene of antibacterial peptide from Tenebrio molitor. All of Tenecin gene and Coleoptericin C gene up-regulate escept Tenecin 3 gene in the larvae of T.molitor induced with E.coli. |
