| Fatty acid is an important component of soybean oil. fatty acid composition and mixture ratio determines the type of seed oil quality, so screening soybean varieties with high fatty acid content and reasonable ratio are one of the important goals in soybean quality breeding. In this study, Two populations of backcross introgression lines derived from the cross of recurrent parent Zhonghuang 13(excellent comprehensive characters) with unrecurrent parent chihuangdou1 and taixingniumaohuangyi(low fat content) were used to map QTL which affects soybean fatty acid component content, we expect to find genes related to oil concentration and fatty acid components, and screen target introgression lines with high oil concentration and reasonable fatty acid components. This can provide the guidance for molecular marker-assisted breeding in soybean. The main results were as follows: 1. Correlation analysis and variance analysis of quality and agronomic traitsThe two populations ZC and ZT existed extensive genetic variation and transgressive segregation between agronomic and quality traits. The oleic acid content shows the widest range of variation and the maximum variation coefficient. The quality traits and agronomic traits of two groups all show significant differences in the population,year, and between family and year, which illustrate five kinds of main fatty acid all are complex quantitative trait, which are jointly affected by genes and the environment. plant height existed significant differences in the family and year, have a a higher degree of variation.which can selected materials with different height types.There were some correlation in the main quality traits, agronomic traits and between quality traits and agronomic traits. Such as oleic acid with linoleic acid and linolenic acid are significantly negative correlation, plant height with bottom pod height and node number are significantly positive correlation and so on. It indicated that there were some stability correlations among the soybean main characters which can provide some references in the coming research. 2. Construction of genetic linkage mapIt screened 166 polymorphic primers between the two parents in the population ZC from 586 pairs of SSR primers which were synthesized in laboratory.The 144 pairs of primers were used for linkage map construction, the total map length was 2569.87 cM, the average genetic distance was 17.85 cM, which cover the 18 linkage groups of soybean except A2 and C1 linkage group. Its screened 176 polymorphic primers between the two parents in the ZT population. The total map length was 2029.46 cM, the average genetic distance is 12.93 cM, which covers the 19 soybean linkage groups except A2 linkage group. 3. mapping QTL related with oil and fatty acidsMapping QTL related with oil concentration and fatty acids components for two populations which was Based on new construction of genetic linkage map.The result was that we located 56 and 71 related QTL respectively. In the ZC group, qSA-B1-1 was repeatedly detected, and markers Sat095-Satt638, Satt309-Sat088 and Sat331-Satt197 were detected with several traits. In the ZT group, markers Sat342-Satt560, Sat322-Sat318 and Satt178-Satt260 were repeatedly detected with the same traits, and the QTL between Satt178-Satt260 was closer together, which may be relatively stable QTL. Markers Sat322-Sat318 was detected QTL which was related to linoleic acid, linolenic and fat. Markers between Sat342-Satt560 was detected QTL relate to linolenic and stearic. |
