Tricholoma lobayense is one species of subphylum Basidiomycotina, Hymenomycetes, Agaricales, Tricholomataceae, Mushroom genus. T. lobayensew which is tasted sweet, delicious, crisp and nutritious, is known as the mushroom flavor both in taste and quality. T. lobayensew is a large hot fungus, its fruiting time mainly in summer and has demanding conditions. However we have fewer high-quality varieties of T. lobayensew, currently only a small amount of the distribution in the coastal areas of Fujian and Taiwan, yet the promotion of inland. To promote the inland areas of T. lobayensew and rich its varieties. Therefore, in this study low-energy nitrogen ion beam were used to induce T. lobayensew and the biological effects of various strains after mutagenesis were studied. At the same time, nutritional value and yield of the mushroom strains were also comparatively studied. And provide a scientific prroff for the breeding study of new quality varieties of T. lobayensew. Moreover, we have starting strains on chitosan coating preservation. By contrast, after treatment of the physicochemical properties of chitosan, we selected efficient and suitable method. By chitosan coating treatment, we can effectively extend the shelf life of T. lobayensew, reduce storage costs, while providing a reference for a composite coating fresh mushroom, with huge economic benefits.Through the experimental results are as follows:1. Through the L9 (34) design of experiment to screen out the optimal mother kind of culture medium formula for T. lobayensew: glucose 25 g/L+peptone 3 g/L+ KH2 PO 42 g/L+potato 200 g/L+agar 20g/L.2. By different doses of low energy nitrogen ion beam injection T. lobayensew strains found mutation strains survival rate after the first reduce with the increase of irradiation dose performance similar to a "saddle type" of increase and decrease of the curve. Induced strain is obtained by low temperature screening JF-004 has the fastest growth, reached 1.168mm/d.3. At the lowest temperature in 20℃, the mushroom for each strain to experiment, the average yield of the JF-003 mutant strain was 185.88±1.93g, biological efficiency of 74.35%; the average yield of JF-004 was 191.60±1.89g, biological efficiency reached to 76.64%. The two mutant strains average yield observably increased than the starting strain. Through the determination of fruiting body of nutrients, JF-004 was screened out because it has the best comprehensive nutritional value, the protein content of 35.26±0.45%, total sugar content of 38.70±0.87%,9.26±0.37% crude fiber, crude fat content was 9.21±0.11%. By tasting the induced strains In the process of experiment, to screen JF-003 which have crisp mouth feeling and higher sweet degree strain, and its high yield and biological efficiency, second only to JF-004.4. Through the L9 (34) design of experiment to screen out the optimal formula of compound coating preservation:chitosan 15 g/L, phytic acid 1.5 g/L, ascorbic acid 1 g/L. Experimental verification of the optimal ratio of the composite coating preservation process by being able to effectively extend the shelf life of T. lobayensew up to 15 days.
