In this paper, two polysaccharide from Tricholoma lobayense were purified. The purified method of Tricholoma lobayense was optimized. The research on structural properties of the two polysaccharides and some biological activity are as follows:1. The optimization of extraction of raw polysaccharides from Tricholoma lobayense. Application of response surface methodology optimized the extraction process of raw polysaccharides. The results showed that the extraction rate of polysaccharides optimum extraction parameters were:extraction temperature 83℃, liquid ratio 1:35, extraction time 2.5h, the actual extraction of polysaccharide rate is 12.294%.2. Two fractions TLH-Ⅰand TLH-Ⅱwas collected after further purified ion exchange and Superdex-200 gel filtration. UV scanning TLH-Ⅰand TLH-Ⅱ, absorption peak are very weak at 260 and 280nm which indicated that they almost do not contain nucleic acid and very little of protein content. Infrared scaned TLH-Ⅰand TLH-Ⅱ, showed polysaccharide compounds have the general characteristic absorption peaks, mainly pyranose. total sugar content of the two components (TLH-Ⅰand TLH-Ⅱ) were 89.6% and 92.3%; protein content were 6.5% and 4.5%; molecular weight were 5.85×105Da and 4.22×103Da. TLH-Ⅱmonosaccharide composition and the molar ratio:arabinose:mannose:glucose:galactose=15.1:2.3:3.7:1.8.3. The experimental model of diabetic mices was induced by alloxan, and the experimental model of hyperlipidemia in mices was established by administration of high-fatty animal feeds for three weeks. During a two-week administration, the levels of TC, TG,HDL-C and blood glucose in diabetic and normal mice were no significant change, and the levels of LDL-C was significantly decreased (P< 0.05). During a one-week administration, the levels of TC, TG, HDL-C and blood glucose in hyperlipidemia mice were no significant change, and the levels of LDL-C was significantly decreased (P<0.05). 4. Tricholoma lobayense polysaccharide can effectively scavenge oxygen free radicals; the production of Hemolysis of red blood cells and MDA in vivo were inhibited; by fluorescence spectroscopy, demonstrate that a certain modification of AAPH oxidation of HSA was inhibited. Oxidation inhibition strength was TLH <TLHⅠ<TLHⅡ.
