Parabronema skrjabini is a parasite which may bring serious harm to the camel breeding industry. In order to understand the prevalence of the Parabronema skrjabini of Camels in Inner Mongolia region in recent years, it is necessary to develop a antemortem diagnostic methode of Parabronema skrjabini of Camels. Firstly, investigation of Parabronema skrjabini of Camels in winter was carried out from 2013 to 2015 in camel main producing area of Inner Mongolia region. Then specific primers of highly specific rDNA ITS region of Parabronema skrjabini were designed, followed by primers’ specificity and sensitivity test with eggs from seven kinds of camels common gastrointestinal nematodes and Parabronema skrjabini. Later, three methods of extracting DNA of Parabronema skrjabini eggs from artificial positive camel dung were used to ascertain antemortem diagnosis method of the Parabronema skrjabini in Camels. The results showed that among 108 camels in main breeding area, the average infection rate of Parabronema skrjabini of Camels was 81.3%, and the highest infection intensity was 1272 worms per camel, while the ratios of the female worms to the male were from 1.7:1 to 2.4:1. The infection rates of camels in Bayannao’er, Wulanchabu, Alashan and Xilin Gol League were 100%,85%,70.9% and 57.1%, repectively. Primers used in this test had advantages of good specificity and high sensitivity, and the fragment of 551bp were got. Besides, it can detect as low as 15 Parabronema skrjabini eggs. Improved methode of CTAB+SiO2 adsorption to extract DNA of Parabronema skrjabini eggs in artificial positive camel dung was used and the detection rate was 80%, which was better than the kit and conventional extraction by saturated phenol-chloroform extraction method. The test’s results identified that the Parabronema skrjabini of Camels remained widespread in Inner Mongolia in recent years, the specific primers and methode of CTAB+SiO2 adsorption can be used in antemortem diagnosis to Parabronema skrjabini, and it laid the foundation for the epidemiology investigation and prevention measures of Parabronema skrjabini in Camels.
