| In this experiment,have used the seeds of Pinus massoniana’s Excellent provenance tongmian as a starting material. After seeds germinated, cut the terminal bud as explants.Then had a series of experiments which included terminal bud elongation growth, adventitious buds induction and elongation growth, induced adventitious root,ect.Have had a series of studies for the tissue culture technology of Pinus massoniana. The test results were as follows:(1)Seed disinfection with 75% alcohol and 0.1% HgCl, only control the concentration of disinfectant and sterilization time could achieve good disinfection. Seeds disinfection with 3% NaClO had a bad effet for seeds germination. Disinfecting seeds without 3% NaClO,the contamination rate was smaller, and higher germination rate.(2) SH medium is the best suited to the terminal bud elongation growth, while the sterile explants terminal bud sprouting which germinated in the culture medium got better elongation growth.(3) There was not much effect on the induction of adventitious buds throug different germination way to get the explants.But the length of induced adventitious buds explants had a certain impact on producing adventitious buds(4) SH medium was the best suited for adventitious buds. And the increasing coefficient of adventitious buds was the max when NAA concentration is Omg/L,6-BA concentration was 1.5mg/L.(5) The best medium which inducted tongmian’s adventitious buds elongation growth was DCR+NAA0.3mg/L+IBA0.8 mg/L.(6) 1/3DCR medium + NAA0.15mg/L + IBAOmg/L was conducive to induce adventitious roots, the induction rate of adventitious roots is the max,which was 76.5%. On the 1st of ABT used in the experiments, the induction of tongmian adventitious roots had no more significant role in promoting.The technology of Pinus massoniana tissue culture became more mature. The above experiment provided a reliable basis for planting Pinus massoniana in the mountain as soon as possible. |
