| In this study, TLR4 gene which is candidate gene about disease resistance traits was selected, its SNPs (single nucleotide polymorphisms) were detected in Wannan Black pig, Anqing Liubai pig, Wei pig, Huo Shou Black pig and landrance, associations between the detected SNPs with immune indexes of weaned piglets in Wannan Black pig, Anqing Liubai pig, Wei pig, Huo Shou Black pig and Large white pig were analysed, to find out the the effective genetic markers of disease resistance traits in Anhui native pigs, and provide reference of molecular biology for the study of germplasm characteristics and MAS in Anhui native pig breeds.The results showed that:(1) TLR4 gene polymorphisms in Wannan black pig, Anqing liubai pig, Wei pig, Huo Shou black pig, and Landrace five were detected, found 3 SNPs located in TLR4 exon 3 were found, namely:G417A, C318A, C1027A.(2) The population genetic analyses showed that:C318A sites in Wannan black pig,wei pig, Anqing liubai pig detected CC, CA genotypes, including genotype frequency in Wannan black pig was 80.95%,19.05% respectively; genotype frequency in the wei pig frequency was 70.00%,30.00%; genotype frequency in anqing liubai pig for 82.50%,17.50%, respectively, CC genotype was the predominant genotype of the three groups, AA genotype was not detected, the polymorphism information content (PIC)of the site in the three groups were 0.1575,0.2225,0.1469, were in the low polymorphism; and through x2 fit test,x2 value of the three groups were less than 5.991, in Hardy-Weinberg equilibrium state (P> (1.05). The loci in Huo Shou and Landrace pig population mutation was not detected.G417A sites in Wannan black pig,wei pig, Anqing liubai pig detected CC, CA genotypes, including genotype frequency in Wannan black pig was 80.95%,19.05% respectively; genotype frequency in the wei pig frequency was 70.00%,30.00%; genotype frequency in anqing liubai pig for 82.50%,17.50%, respectively, GG genotype was predominant genotype of the three groups, AA genotype was not detected. The loci in Huo Shou and Landrace pig population mutation was not detected.C1027A sites in five herds were detected CC, CA, AA genotype, which genotype frequency in Wannan black pig 23.81% 52.38%,23.81% respectively; gene in the wei pig genetype frequency was 40.00%,35.00%,25.00%; genotype frequency in anqing liubai pig was 27.50%,45.00%,27.50%; genotype frequency in Huo Shou pig was 6.98%, respectively,65.12%,27.91%; genotype frequencies in the Landrace was 59.09%, 27.27%,13.64%.The PIC value of five groups were 0.3750,0.3693,0.3750,0.3683,0.3180 both in moderate polymorphism; and after x2 fit testing, x2 values of five populations are less than 5.991, in Hardy-Weinberg equilibrium (P> 0.05).(3) Blood immune indexes in group the differences between the analysis:The test using enzyme-linked immunosorbent assay kit for Anqing liubai pig, Wei pig, Huoshou black pig, Large White pigs weaned four interleukin-6 (IL-6), interleukin-10 (IL-10), interleukin-12 (IL-12), y interferon (IFN-y), tumor necrosis factor-a (TNF-a)) five immune parameters were determined for each immunization indicators showed large differences between the different levels of breeds; in addition to the level of IL-6 and IL-10 levels are not significant differences in the wei pig and large White, IL-12 levels in Huoshou black pig and large White were not significant, the differences between various other immune indicator species are extremely significant.(3) With the immune indexes of correlation analysisThe test also analyzed C1027A sites and G417A polymorphism in Anqing liubai pig, Wei pig, Huoshou black pig, Large White pigs weaned four of five immune parameters (IL-6, IL-10, IL-12, IFN-y, TNF-a) correlation:for C1027A site only found that Wei pig TNF-a levels AA genotype were significantly higher than CC and CA genotype; large White IL-10 levels CC genotype were significantly higher than those of CA genotype,.The study suggested that:C1027A loci of TLR4 gene shoud be helpfulto be used as a targeted point in anhui local pig breeds, anqing liubai pig,wei pig, HuoShou black pig and large white pig disease resistance breeding. |
