The unrooted tissue culture seedlings of Musa cavendishii and Eucalyptus were used to study effects of different intensity and color of LEDs, ratio of red and blue LEDs, lids of tissue culture container, different medium matrix on their growth. The results are as follows:1、Red light promoted the elongation of the root of tissue culture seedlings of Musa cavendishii.with the increase of intensity of red light, the dry matter of aboveground, the content of carotinoid and total soluble sugar in shoot also increased, but the dry matter of root declined; excessive blue light inhibitted accumulation the seedling height, but promoted the accumulation of total soluble sugar in leaf.2、under 765 Lx and 80% R(red led)+20% B(blue led), root number, root length, plant height, stem diameter, root weight, total seedling weight and seedling weight-increasement of the tissue culture seedlings of Musa cavendishii were the most, and signficiantly higher than the fluorescence treatment except for root length; under 765 Lx, 40% R+60% B, the leaf number, leaf area were the highest, and signficiantly higher than fluorescence treatment; under 765 Lx, 60% R+40% B, weight of aboveground, content of total soluble sugar of stem were the most; under 765 Lx, 20% R+80% B, dry matter content of root, contents of total dry matter in seedling were the most; under 612 Lx, 25% R+75% B, dry matter content in aboveground, chlorophyll a/b ration were the most; under 459 Lx, 67% R+33% B, contents of chlorophyll a and chlorophyll b were the most.3、when common lid was used, root length, plant height, weight of aboveground, weight of root, weight of whole seedling, weight increasement, and content of total soluble sugar of the tissue culture seedlings of Musa cavendishii were the most, and were significantly higher than other two lids; using common lid showed no significant difference in root number, leaf area and stem diameter from other treatments; when breathable film was used, leaf number was the most; when breathable lid was used, content of dry matter in root, contents of chlorophyll a, chlorophyll a+b, and chlorophyll a / b ratio were the highest, and significantly higher than other two lids.4、when cultured on agar medium, root length, plant height, leaf number, leaf area, stem diameter, aboveground weight, root weight, whole seedling weight, contents of dry matters of aboveground, contents of chlorophyll a, chlorophyll b, carotenoids and chlorophyll a + b and contents of total soluble sugar of the tissue culture seedlings of Musa cavendishii were significantly higher than the other two media; Under fluorescent lighting, content of dry matter in root cultured on coco coir medium and vermiculite medium, were significantly higher than on agar medium.5、red light promoted increasing of leaf area; blue light promoted elongation of root, increasing of plant height, increasing of root weight in the cultivatiom of tissue culture seedlings of Eucalyptus; with increasing of intensity of red light, number of root, leaf area, content of chlorophyll a also increased; with increasing of light intensity of blue light, root length, plant height, leaf number, leaf area, root weight also increased; excessive blue light inhibitted formation of contents of total soluble sugar in leaf, but excessive red light promoted formation of contents of total soluble sugar in leaf; under the same intensity of different light quality, chlorophyll a/b ratio was higher under blue light than under red light, and the contents of total soluble sugar in leaf was higher under red light than under blue light.6、In the cultivatiom of tissue culture seedlings of Eucalyptus, under 765 Lx, 80% R+20% B, root number and root length were the most, and seedling height was second among all treatments, but showed no difference on leaf number and leaf area from the highest treatment; treatment of 765 Lx and 80% R+20% B showed no significant difference in weight of aboveground, whole seedling weight, contents of dry matter of aboveground from the highest treatment. Under 765 Lx and 60% R+40% B, root weight, total weight were the highest among all treatments; under 459 Lx, 67% R+33% B, dry weight of aboveground were the highest among all treatments; under 612 Lx, 75% R+25% B, content of chlorophyll a, chlorophyll b and carotioids were the highest among all treatments; under 612 Lx, 75% R+25% B, contents of chlorophyll a, chlorophyll b and carotioid were the highest among all treatments, but chlorophyll a/b ratio were the highest; under 612 Lx, contents of total soluble sugar in shoot and root were the highest, and lowest under 765 Lx.7、when common lid was used, root number, root length, plant height, leaf number, shoot weight, root weight, plant weight and chlorophyll a / b ratio of tissue culture seedlings of Eucalyptus were significantly higher than the other two treatments; when breathable film was used, formation of chlorophyll a, carotinoid, chlorophyll a + b were promoted, and leaf number was the biggest among all treatments; when breathable lid was used, formation of total soluble sugar was promoted.8、Unrooted tissue culture seedling of Eucalyptus was used to study effects of different medium matrix on their growth. Results indicated that: rooting rate was almost 0% when cultured on coco coir medium; when cultured on agar medium, root number, root length, plant height, leaf number, content of all kinds of chlorophyll and carotinoid were significantly higher than the other two treatments.
