Picea mongolica is a rare tree species endemic to Inner Mongolia.In this experiment,the mature embryo as explant materials of Picea mongolica,Study on the influence of different culture media, different hormone combinations, different culture conditions to embryogenic callus induction and proliferation,somatic embryo maturation and germination of Picea mongolica.Finally,the system of somatic embryogenesis of Picea mongolica is established.The main conclusions are as follows:1.It determines the best combination of embryogenic callus proliferation culture of Picea mongolica is LM+6mg/L 2,4-D,2mg/L 6-BA and 0.5mg/L KT. with 500 mg/L casein hydrolysate,450 mg/L glutamine,20g/L sucrose and 3g/L Gelzan gelatin, adjust the pH value to 5.9.The optimum culture conditions is dark culture,and the rate of embryonic callus induction is 46%.2.It determines the best combination of embryogenic callus proliferation culture of Picea mongolica is LM+O.lmg/L NAA,0.5mg/L 6-BA,0.2mg/L KT.Additional organic additives such as induction medium’s.the best proliferation effect is 4 times of subculture. And multiplied 2.72 times.3.It determines the best combination of somatic embryo maturation culture of Picea mongolica is LM+5.0mg/L ABA,0.5mg/L IBA,5.0mg/LAgNO3.with 500 mg/L casein hydrolysate,450 mg/L glutamine,1g/L inositol,40g/L sucrose and 3g/L Gelzan gelatin, And the two generation (40d) later, a mature somatic embryos.The rate of somatic embryo maturation is 88%.4.It determines yhe best combination of somatic embryo germination culture of Picea mongolica is LM medium with lg/L activated carbon 500 mg/L casein hydrolysate,450 mg/L glutamine,30g/L sucrose and 3g/L Gelzan gelatin, without any hormone.The culture conditions is 1/2 light and dark,the intensity of illumination is 12001x.The rate of somatic embryo germination is 40%.
