Dairy cattle endometritis is one of the major causes of infertility in dairy cows, which restricted the development of dairy industry in China. Its high incidence, making China’s diary industry suffered a great loss. The most frequently used treatment of endometritis in dairy cows are intrauterine perfusion and flushing method inside the uterus, and the commonly used drugs include antibiotics and chemical drugs. But in recent years, the overuse of antibiotics and chemical drugs have caused drug residue problem. These problems have being reported more, and the overuse of antibiotics leads to drug-resistant strains quantity and speed increasing. How to find new organic and affordable medicine which can cure milk cow endometritis better has become an urgent problem.Mongolian medicine is one of the four traditional Chinese medicine in China. Mongolian medicine has the charactors as used simply, low consumption, and better effect. Therefore this study screen Mongolian herb medicine from Mongolian medical Pharmacopoeia which has antibacterial and anti-inflammatory activity,through through modern research methods, and hope filtering out the drug prescription from these Mongolian herb medicine which can cure milk cow endometritis effectively. This has great significance for milk cow endometritis treatment in China.Main research contents and conclusions are as follows:1.The main pathogenic bacteria were isolated, purificated, and biochemical identificated from cow uterine secretions by regular methods. The main pathogenic bacteria were Escherichia coli and Staphylococcus aureus, and achieved 64 strains of Escherichia coli,17 strains of Staphylococcus aureus;2.Through the test of anti-"o" serotype on 64 strains of pathogenic Escherichia coli which were isolated from cow uterine secretions,33 strains of pathogenic Escherichia coli and 11 serotypes had been isolated, which are O86, O1, O126, O111, O29, O28,O8, O91, O2, O26, O25. The separation rate of O86 is up to 14%, which makes O86 be the dominant serotypes. There are 4 strains had self-curing phenomenon, and the remaining 27 strains were failed to be detected.3.Combined orthogonal method designing experiment programme, through the antibacterial activity in vitro,4 Mongolian herb medicine compound has been flitered out after compound components filter and dose optimization. They are compound Ⅰ:rhubarb 70g, and Salvia 50g; compound Ⅱ:scutellaria 70g, and rhubarb 50g; compound Ⅲ: rhubarb, and Myrobalan each 50g, Forsythia 30g; compound V:scutellaria, and rhubarb, and Myrobalan each 70g;4.The results of protection rate test of compound Mongolian herb medicine on infected mice by 80% lethal dose are as follows:compound Ⅰ, Ⅱ both are 40%.Compound Ⅲ are 60%. compound Ⅴ are 50%, the effection of compound Ⅴ is equivalent to Ciprofloxacin gatifloxacin,and the effection of compound III are better than Ciprofloxacin gatifloxacin.5.The antibacterial spectrum of the Mongolian medicine be tested in vivo. Streptococcus agalactiae, Salmonella typhimurium, Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus cereus has been tested.The reuslts are as follows: All the herb medicine compounds are highly sensitive or extremely sensitive to the 6 standard strains. And the inhibition zone diameters of compound II significantly higher than other compounds (P<0.05);6.The effects of Mongolian herb medicine compound on the growth curve of the two pathogenic bacteria were determined by turbidimetry. Moreover, the effects of Mongolian herb medicine compound on the hydrophobicity of the the two pathogenic bacteria cell surface were determined using the microbial adhesion to solvents method, and the permeation of the two pathogenic bacteria Cell membrane were determined by measuring the release of β-galactosidase activity into the culture medium using ONPG as a substrate;the effecs of Mongolian herb medicine compound on the two pathogenic bacteria cell wall were determinated by Determination of alkaline phosphatase (APK).The results are as follows:the 4 Mongolian medicine compound all could affect the growth curve of bacteria well, and can affect cell surface hydrophobicity, cell membrane permeability and cell walls from varying degrees.
