It is very important to accurately identify the exogenous chromosome fragmentsduring creating the translocation lines. To achieve the precise identification of thecabbage chromosome fragments in Chinese cabbage-cabbage translocation lines, a totalof91SSR markers and458InDel markers derived from9linkage groups of cabbagewere screened on four cabbage and four Chinese cabbage varieties by PCR amplification.The result showed that320markers were specific on cabbage compared with Chinesecabbage. One Chinese cabbage-cabbage translocation line developed by our laboratorywas identified preliminarily by specific InDel markers on linkage group C02. The mainresults are shown as follows:1. A total of91SSR markers and458InDel markers derived from9linkage groupsof cabbage were screened on four cabbage’11-1’,‘8398’,‘Lan Bao’ and ‘Wan Fengcabbage’ and four Chinese cabbage ‘85-1’,‘Xin Jinlv’,‘Beijing Xinsanhao’ and ‘QiuZhenbai’ varieties by PCR amplification,320specific molecular markers on cabbagecompared with Chinese cabbage were identified. The result showed that34SSR markerswere specific on cabbage compared with Chinese cabbage, which were distributed on9linkage groups of cabbage,1of them on linkage group C01,5of them on linkage groupC02,4of them on linkage group C03,2of them on linkage group C04,2of them onlinkage group C05,3of them on linkage group C06,3of them on linkage group C07,7of them on linkage group C08, and7of them on linkage group C09. And286InDelmarkers were specific on cabbage compared with Chinese cabbage, distributing on9linkage groups of cabbage,31of them on linkage group C01,45of them on linkagegroup C02,54of them on linkage group C03,24of them on linkage group C04,22ofthem on linkage group C05,20of them on linkage group C06,32of them on linkagegroup C07,34of them on linkage group C08, and24of them on linkage group C09. Asystem of specific InDel markers on cabbage compared with Chinese cabbage wasinitially established.2. One Chinese cabbage-cabbage translocation line, AT7-13, containing fragmentfrom C02linkage groups of cabbage developed by our laboratory was identified preliminarily by45specific InDel markers on linkage group C02. Only6plants hadcabbage specific bands on specific InDel markers C02-45(physical initiation sites18551678bp) in the35F2individuals of AT7-13selfing. But there was no cabbagespecific bands between flanking specific InDel markers C02-14(physical initiation sites18096496bp) and C02-15(physical initiation sites18786494bp), indicating that theintroduced fragment for translocation line was located between those two markers.3. Meiosis of6F2plants of AT7-13selfing with specific bands of cabbage wasobserved. The results showed that during the process of meiosis chromosomes hadnormally federation, without univalent and multivalent, suggesting that this six plantshad cytogenetic stability.4. By using specific InDel markers C02-45,18plants with specific markers ofcabbage were identified from19F3plants derived from selfing of AT7-13....
