On the basis of a wide range of materials of Alpinia oxyphylla Miq., the paper will analyzes the genetic diversity of A. oxyphylla Miq. From morphological characters, agronomic characters and ISSR DNA markers, and select several high grade A. Oxyphylla accessions as original material for seeking and conservation of new varieties in the future. The paper will study the Seed Germination and Storage Characteristics with different chemical treatments and different storage conditions. The purpose of this study is to provide some references for the further selective breeding, protection and study on A. oxyphylla Miq. Germplasm resource.1. The traits of flower, fruit and blade of A oxyphylla have a certain variation, most of the traits have obvious difference in different individual plant but are all the same in one. The length of the infructescence, the fruit size, the fruit shape have a larger variation and are closely related with yield, so, they can be regarded as the most important characters for distinguishing and identifying the germplasm resources of A. oxyphylla. The agronomic characters have a larger variation than morphological characters, and that will be useful for breeding program of A. Oxyphylla.2. Correlation analysis showed that The degree of shadowing have a significant impact on the growth of A. oxyphylla; It is significantly and positively correlated between most of parts of a flower, The length of the infructescence is significantly and positively correlated with The length of labellum, calyx tube and middle-lobation are significantly and positively correlated with the fruit size, The length of leaf is significantly and positively correlated with The width of flower, The width of leaf is significantly and positively correlated with the diameter of stem, The length of calyx tube and the width of labellum. The result of Principal Component Analysis model shows:The cumulative variance proportion of the top seven principal components is89.29%, that could contain all traits and can be summed as:the fruit size factor, the yield factor, the fruit shape factor and the vegetative organs factor. The rusults of cluster analysis based on phenotypic characters classified72samples into9groups.3. The results of ISSR of A. oxyphylla germplasm show a total of466fragments were detected,446fragments were polymorphic, accounting for95.71%of the total. The genetic similar coefficient of68A. oxyphylla germplasm was0.72-0.86, that indicate a low levels of genetic diversity.4. The seed germination of A. oxyphylla was slowly and lasted a long period in normal, and can be improvement with different chemical treatments such as GA3(300-700mg/L) and NAA(25mg/L).The seed could not be stored for a long time at room temperature that its germination rate would decreased quickly to below50%after40days and to0%after90days. The seed also could not endure high temperature that higher than60℃and could not endure water loss exposed to the sun one day or in a silica dryer.
