Seed orchard served as the bridge of breeding between the forest breeding and forest production is the most important population of seed production. The genetic diversity will directly affect their stability and adaptability of forest population.Clones in seed orchard of P. massoniona in the Institution of Shaoguan Forestry Science, Guangdong Province were studied by both morphological markers and RAPD markers.The genetic diversity were also conducted.The diameter of stem, height of stem, and stem volume, 100 cone weight, thickness of cortex and linear-leaf length of 10 families were recorded. The statistics of morphological characters with Means, ANOVA and Coefficient of Variance showed that abundant genetic variance was found within families and among families of 6 morphological characters.The Cluster analysis by software NTSYS2.10 showed that all the mason pine families could be divided into three types: the southern of guangdong province type,the northeast of guangdong province type and the middle of guangdong province type.Sixteen primers of 10 base pair were screened out from 210 random primers on the genomic DNA of P. massoniona with RAPD technique.And generated 118 bands,of which polymorphic bands number was 95, the percentage of polymorphism equaled 80.37%. As analyzed by software POPGENE, the genetic diversity of P. massoniona was relatively high (H_e=0.2445; H_o=0.3558).And comparison of genetic diversity in other species in the Coniferopsida indicated that the genetic diversity of P. massoniona was higher Iever.The main results were as follows:1. A rapid and simple DNA extraction method for RAPD analyses from the pine needles was reported.And the improved CTAB subarea method was used for the first time for genomic DNA isolation from pine needle.2.The optimal reaction system was structured by studying the factors affecting the pine RAPD reaction.3. The statistics of morphological characters showed that the GL population was the most rapid growth,.It should pay attention to in the forest breeding.4.The analysis of morphological markers and RAPD markers showed that the geneticdiversity within families was larger than the genetic diversity among families in clones of P. massoniona in seed orchard. The cluster analysis proved that all families could be divided into three types: the southern of guangdong province type,the northeast of guangdong province type and the middle of guangdong province type.5. The analysis of morphological markers and RAPD markers indicated that clones of P.massoniona in seed orchard were larg genetic diversity. This seed orchard had an ability to product good genetic trait ,high genetic diversity and stability of forest population and satisfied the forest production in the forest breeding.
