Agrobacterium-mediated immature embryo transformation is often used in maize. This method has higher transformation efficiency, but it depends heavily on genotype, and the immature embryos are limited as a result of seasons.In order to establish a simple and easy maize genetic transformation system, the Agrobacterium-mediated genetic transformation of minimally invasive maize shoot tip was studied. In the experiment, the maize inbred lines Zheng58,2193,9002were used to explore the effect of different Agrobacterium infection time and different vacuum pressures on maize regeneration rate via Agrobacterium-mediated transformation. The results were as follows:1. The regeneration rate of Zheng58is highest in three maize inbred lines under30minute Agrobacterium infection, and with vacuum pressure increases, plant regeneration rates gradually reduced.2.3transgenic maize plants were primarily identified using PCR method.3. The method of Agrobacterium-mediated genetic transformation of minimally invasive maize shoot tip has a simple experimental procedure and short testing process because the co-culture stage is not afraid of pollution and the new leaves grow faster due to the retention of maize root.
