Rice was attacked by a serious invasion of pests and diseases in the production. In recent decades, ordinary rice dwarf disease was highly occurred in some regions of China, causing serious losses. In2014, China’s ministry of agriculture awarded security certificate for transgenic crylAb/crylAc gene rice (Huahui1and Shanyou63) and allowed the application in Hubei province for five years (2014-2019). This indicates that China is likely to become the first country to carry out the commercialization of transgenic Bt rice cultivation. So, we want to determine what is the relationship between transgenic Bt rice-rice dwarf virus-vector insect? Whether widely cultivated transgenic Bt rice can cause serious occurrence of rice diseases and insect pests? In order to solve the mystery, experiment was carried out around the rice dwarf disease. The disease resistance level of two types of transgenic Bt rice, transgenic cry1C gene rice (T1C-19) and transgenic cry2Agene rice (T2A-1), and their effect on biological parameters of green rice leafhopper (Nephotettix cincticepswere) conducted with their non-transgenic parental plants(MH63), aiming to judge the ecological safety of transgenic Bt rice, and then providing a scientific basis for the success commercialization of transgenic rice; In addition, the biological characteristics of single female leafhopper homozygous lines and the ability of vector acquiring RDV among different generation were researched, exploring the reasons for why homozygous lines leafhopper populations was difficult to maintain and the change trends of virus acquisition ability with heterozygosity.1The resistance of transgenic Bt rice to rice dwarf disease.The resistance of Btrice lines (T1C-19and T2A-1) to RDV under no-selection conditions were compared with their non-transgenic parental plants MH63. Results indicated that two transgenic Bt rice lines were more susceptible, Infective Rate and Disease Index were significantly higher than that in non-transgenic parental plants, but there was no significant difference between two transgenic Bt rice lines. The resistance of Bt rice lines (T1C-19and T2A-1) to RDV under selection conditions were also analyzed. T2A-1performed the highest disease resistance level; Infective Rate and Disease Index were significantly lower than two other lines.Feeding preference bioassay indicated that infected-leafhopper showed different directional response to the three kinds of rice lines. Feeding preference of infected-leafhopper were no significant difference among three rice lines at the initial phase after injected insects (2h), but as the extension of feeding time, T2A-1and MH63showed greater attractive than T1C-19significantly, more insects were loading and feeding on them, but no significant difference between T2A-1and MH63.2Effect of infected-transgenicBtrice on biological parameters of leafhopperFor transgenic/non-transgenic rice plants:compared with the parent line MH63, transgenic Btrice plants showed great effect on some biological parameters of the GRL.T1C-19rice did not markedly influence most of the biological parameters although egg duration, average fecundity and fresh weight of newly emerged adult decreased significantly, nymphal duration increased significantly. T2A-1markedly decreased theegg duration, average fecundity and fresh weight of newly emerged male adult, nymphal duration and fresh weight of newly emerged female adult increased significantly.For infected/healthy rice plants:RDV also affecting the biological parameters of the GRL.Compared with the healthy MH63, egg duration, nymphal survival rate, fresh weight of newly emerged female adult, life span of male adult and average fecundity were all increased significantly on infected-MH63rice plants; Infected T1C-19rice markedly influence most of the biological parametersincluding nymph survival rate, fresh weight of newly emerged female adult, life span of male adult and average fecundity were all increased significantlywhile nymphal duration decreased significantly compared with healthy T1C-19; Infected T2A-1also showed a similar performance, after infected by the RDV virus, egg duration, nymphal survival rate life span of male adult and average fecundity were all increased significantly, fresh weight of newly emerged female adult markedly decreased, other biological parameters showed no significant difference.Most of the biological parameters of GRL were not significantly affect by transgenic/non-transgenic rice plants X infected/healthy rice plants interaction, only significantly prolong life span of male adult.3Biological parameters and virus acquisition ability of single female leafhopper homozygous linesPopulation quantity of leafhopper was decreased as the heterozygosity decrease,significantly reduced egg hatchability and nymphal survival rate were the main factors, but other biological parametersshow no marked difference.Virus acquisition ability of leafhopper was decreased as the heterozygosity decrease.
