In this paper, the pure culture method was used to carry out bacterial culture of the plant(Populus),and by morphological and molecular biology to determine the composition of the root and the rhizosphere bacterial communities of the poplar.;By comparing healthy plants with non-healthy plants(Poplar with Botryosphaeria dothidea) and changes in the composition of roots and rhizosphere bacterial populations,we can clear the influence on the community structure of Poplar with Botryosphaeria dothidea,and it can provide an important theoretical basis for the pathogenesis;And comparing the differences in bacterial composition of Populus roots, rhizosphere and other relatively resistant, it can provide a theoretical foundation for the relationship between plant resistance and microbial diversity.The following main results obtained in this study :The diluted coating method was used to diluted coating the samples on R2 A and NA solid medium. Root endophytic bacteria biomass was found in the range of 103 to 104, bacterial biomass in the rhizosphere and non rhizosphere soil was found in the range of 105 to 106,and the root of the whole circle of soil bacterial biomass on less than half of the rhizosphere soil bacteria biomass.In different health of samples,the overall health of the plants is less than the non-healthy bacteria.In different resistance samples,the plant resistance is stronger, the less biomass in the root endophytes,and the rhizosphere and the non-rhizosphere biomass of soil bacteria is substantially the same.At the same time,we found Poplar endophytic bacteria biomass in the root and bacterial biomass in the rhizosphere soil was positively correlated.The stains’ 16 S rDNA sequencing results showed that it belongs to 27 genera,and 76 species.Among them, the health of plant samples obtained 22 genera, a total of 53 species; nonhealth(with poplar canker) samples obtained 22 genera,46 kinds. Roots isolated endophytic bacteria belonging to 16 genera and 34 species;rhizosphere soil bacteria isolated belonged to 13 genera and 39 species; non-rhizosphere isolated 16 genera, 31 species.Pseudomonas and Bacillus for the advantages of all sample collection genus,which each accounted for 33%,15%.At the same time, in root samples,Staphylococcus account for a large proportion of 8.8%; In the rhizosphere samples, Lysobacter and Ensifer account for a large proportion of 7.7% and 7.7%; In the non-rhizosphere samples, Agromyces and Streptomyces account for a large proportion of 6.2% and 6.2%.
