| This experiment aimed to investigate the influence factors and mechanism of regulating expression of MP1 and GMPR2 genes,and the formation and development of hair follicle were prospected to understand, which provided constructive advice and help for further breeding Liaoning cashmere goats. First of all, collecting the skin to cell culture, then the specific lentiviral RNAi expression vector targeting Noggin gene was constructed, which infected cells after packaging. Otherwise, the exogenous cytokines(fibroblasts growth factors5 and insulin-like growth factor1) and melatonin were used to treat goat fibroblasts cells by both single and joint. Finally the expression quantity of MP1 and GMPR2 gene was calculated and analyzed by Thermal Cycler DiceTM System Software.The results were as follow:(1)The best value of MOI of Lentivirus vector was 30, and which needed polybrene to enhance the efficiency of infection.(2)Successful building the Lenti- Noggin- EGFP- miR and calculating the virus titers, which was 1.35×106Tu/ml.(3)The expression of GMPR2 was the lowest in the group of Noggin RNAi, at the same time, the highest was the group of NC.(4)The results of fibroblast cells treated by Melatonin showed the expression of MP1 gene was inhibited, but with prolong the inhibitory effect was weakened.(5) The expression of MP1 and GMPR2 genes were very significant difference in any period by FGF5 treating fibroblasts, respectively the maximum value appear in the 10-6g/L of 24 hours and 10-6g/L of 72 hours.(6)The inhibition of expression of MP1 gene was decreased gradually with drug treatment’s time prolong by fibroblasts treated by IGF-1. While the maximum value of GMPR2 gene appeared in 10-4g/L of 72 hours.(7)The experiments proved that MT combine with either kind of drug can promote the expression of two genes. While the effect of combining with FGF5 was more obvious.In conclusion:(1)BMP signaling pathways played a negative regulation role in expression of GMPR2 gene.(2)Melatonin has a positive relation of regulatory effect on FGF5 and IGF-1. The combination can promote the expression of the two genes. Which provides support of data for further study the mechanism of cytokines and melatonin above gene expression, then provide a theoretical basis of the effect on fibre traits. |
