| Liaoning cashmere goats,which yield large amounts of high-quality cashmere wool,are among China’s most precious genetic resources.A new gene called Ovarian cancer immuno-reactive antigen domain-containing protein 2(OCIAD2)which was found in skin of Liaoning cashmere goat by our laboratory lab.OCIAD2 is related to the cell cycle of hair follicles and hair follicle development.Melatonin(MT),fibroblast growth factor 5(FGF5)and insulin-like growth factors-1(IGF-1)has come up a lot in the field of cashmere goats,which are closely related to cell cycle changes of hair follicles and the regulation of follicle development.The experiment’s aim is to detect the changes of OCIAD2 expression in skin cells and cell proliferation by infecting cells with MT,FGF5 and IGF-1.Addition,infecting cells by simultaneous treatment with fibroblast growth factor 5 or IGF-1 and MT,and detecting changes of OCIAD2 expression in skin cells and cell proliferation.Methods include Primary cells culture,MT,FGF5 and IGF-1 action cells,RNA extraction,Reverse transcription,RT-PCR,QRT-PCR,CCK8 and detect cell cycle by flow cytometry instrument.After a serious of experiments,the results are shown as follows:(1)OCIAD2 is significantly decreased by MT and FGF5.OCIAD2 is decreased by IGF-1 for 24 h.OCIAD2 is significantly decreased by the concentration group with FGF5 and MT,but significantly increased by the simultaneous concentration group with IGF-1 and MT.(2)Cell proliferation is inhibited by MT.There is no significant effect by FGF5 and IGF-1.Simultaneous concentration group with MT and FGF5 inhibit the cell proliferation.Simultaneous concentration group with MT and IGF-1 inhibit the cell proliferation too.Conclusion:(1)Both of MT and IGF-1 could down-regulated the expression of OCIAD2 through TGF-β singnaling pathway,while FGF5 could up-regulated the expression of OCIAD2.Simultaneous concentration group with MT and FGF5 could down-regulated the expression of OCIAD2.That means the inhibition of MT is stronger than FGF5’s.However,Simultaneous concentration group with MT and IGF-1 could up –regulated the expression of OCIAD2.That means MT,IGF-1 and OCIAD2 should have some kind of signal transduction,and OCIAD2 may be involved in the regulation of the hair follicle with IGF-1 and MT.(2)MT inhibited the proliferation of cells,FGF5 and IGF-1 had no significant effect on the proliferation of cells.But two synergistic groups of MT and FGF5,MT and IGF-1 still inhibits the proliferation of skin cells,which was inhibited to a greater extent than alone,indicating that MT plays the dominant one.The OCIAD2 expression may have a positive correlation with cell proliferation. |
