As a vegetative propagation crop, potato’s genetic is gradual depression by infected with virus after cultivating several years, which lead to the losses of production and quality. At present, the effective way to solve this problem is using virus-free pre-basic seed potatoes (microtubers) in cultivation. To develop a substrate with rich source and good quality for increasing yield of potato microtubers and utilizing agricultural waste reasonably, the experiment analyzed and optimized the ratio among mushroom residue, sand and perlite. The results showed as follows:1、 Quadratic-orthogonal-rotation-combination design was adopted in this experiment, the regression model was established between yield of microtubers (y) and mushroom residue(xi), sand(x2) and perlite(x3), which was expressed as:y=36.33163-1.81965x1+3.39104x2+2.56847x3-1.78637x12-4.20821x32+2.12250x1x2+4.32750xix3.The results showed the influence of three components to the yield of potato microtubers was decreased in the order of mushroom residue, perlite and sand. It was also concluded from the model that there was no significant interaction between sand and perlite to the yield, while significant interaction between mushroom residue and sand become obviously, the same was true between mushroom residue and perlite. The volume ratio of mushroom residue, sand and perlite was optimized as follows:43.03%:30.53%:26.44%.2、{3,3} simplex lattice design in the mixture experiment design was adopted in this experiment, the regression models about high yield and good quality were established to optimize the ratio of mushroom residue, sand and perlite. when obtained high yield of potato microtubers, the volume ratio of mushroom residue, sand and perlite was optimized as follows:46.77%:31.94%:21.29%; when obtained great number of potato microtubers, the volume ratio of three components was optimized as follows:45.00%:29.47%:25.53%; when obtained high weight of potato microtuber, the volume ratio of three components was optimized as follows:45.41%:26.76%:27.84%; when obtained high rate of large and medium-sized microtubers, the volume ratio of three components was optimized as follows:45.24%:31.90%:22.86%. Overall consideration of the above indexes, when obtained high yield and good quality of potato microtubers, the volume ratio of three components was optimized as follows:45.60%:30.02%:24.38%.
