Panax ginseng C.A.Meyer is the rare medicinal plant, and is one of the major specialties in Jilin Province.Changbai Mountains become main producing areas of ginseng with its unique geographical location and climatic condition in Jilin Province. P. ginseng belongs to the highly bogey plant, there is a continuous cropping obstacles, long-term use of deforestation to plant ginseng, affect sustainable development. Therefore, the development of P. ginseng industry has been hampered, P. ginseng continuous cropping must be resolved as soon as possible. Most scholars believe that the continuous cropping obstacles and deterioration of soil physical and chemical properties, there is a close relationship between changes in microbial systems. However, soil improvement can only relieve continuous cropping symptoms in a certain extent, there is also have the occurrence of disease, and low yield problem. The experiment was analyzed from the perspective of rhizosphere microorganisms. Use of microbial pure culture, PCR-DGGE technique, gene cloning methods to carry out continuous cropping P. ginseng research question, Focus on the impact of fungi on plant diseases. Provide a basic disease prevention of P. ginseng disease prevention, pathology research and the development of microbicides. The main conclusions of the study are as follows:1、 Established P. ginseng rhizosphere soil fungi ITS region PCR reaction system, the best results is 25μL reaction system DNA concentration 52.5 ng, dNTPs concentration of 0.20 mM, Mg2+ concentration of 1.50 mM,4.8μmol primer concentration.2、 Screening for fungal DGGE fingerprint analysis primer ITS1/ITS4, ITS1-GC/ITS2, fungi DGGE gels and determining the monomer concentration of 12% to 6%, denaturant concentration of 60%-40%. Bacterial analysis using primers 314F-GC/518R, DGGE gel monomer concentration of 8%, denaturant concentration of 60%-40%.3、 The results showed that rhizosphere and non-rhizosphere fungi significantly different fungal species, and fungal species with increasing planting years becomes less. With the increase in planting years in the number of the same species of fungi have changed, the trend showed fewer, and there are a few new fungal growth, P. ginseng rhizosphere bacterial and non-rhizosphere bacterial diversity index did not change significantly, but there is much more than the reference to the amount of bacteria without reference, acquired breed bacteria patliogens may existed.4、 By cloning sequencing has obtained 68 sequencing results, analysis the sequencing results of system, comparison against ginseng rhizosphere soil were mycorrhizal fungi ginseng rhizosphere soil were Mycorrhizal fungus、 Basidiomycota sp.、 Euascomycete sp.、 Euascomycete sp、 Mortierella alpina、 Ascomycete sp.、 Geomyces sp.、 Paraglomus laccatum、Cryptococcus podzolicus and Ascomycota sp..
