Effectiveness And Methods For Quantitative Determination Of Peracetic Acid Glutaraldehyde Compound Disinfectant

In order to further explore the effect of Peracetic acid glutaraldehyde compound disinfectant on Bacillus, oocysts and parvovirus and its quality control method, we determined to confirm the reliability of effectiveness and method of application by testing effectiveness of Peracetic acid glutaraldehyde compound disinfectant against Bacillus subtilis var. niger, Eimeria tenella, and PPV(porcine pavrovirus virus), combined with researches on active ingredient content of the disinfectant by HPLC (High Performance Liquid Chromatography). It is also tested whether it is specificity, repeatability, accuracy, linear dependence or LOD(limit of detection). The results showed that:(1) It is conducted by quantitative and qualitative suspension tests to determine the disinfection effectiveness of Peracetic acid glutaraldehyde compound disinfectant(under different dilution ratio and action time against Bacillus subtilis var.niger). The result showed that it can kill the spores of Bacillus subtilis var. niger effectively under the conditions of 2~64 times dilution and 5 min, or 256 times dilution and 30 min; In addition, the killing rate can reach 100% via 256 times dilution and 45 min, or 64 times dilution and 7.5 min. It demonstrated that the MBC (Minimal bactericidal concentration) of Peracetic acid glutaraldehyde compound disinfectant against spores of Bacillus subtilis var. niger was 0.39%, with the shortest time sterilization 5min.(2) By animal test (SPF chicken) to determined the disinfection effectiveness of Peracetic acid glutaraldehyde compound disinfectant (with different dilution ratio and action time against oocyst of Eimeria tenella). Which turned out that, sporulated ratio of immature oocyst declined unapparent under 8-2048 times dilution,5-60min; as concentration and working time increase, counting of chicken caecum lesion, productivity of oocyst, dynamiting depletion rate, conducted by oocyst of sporulating Eimeria tenella declined. However, ACI(anti-coccidial index aci) was below 160, which means that oocyst of Eimeria tenella cannot be exterminated effectively.(3) By PK15 cells infection test to determined the disinfection effectiveness of Peracetic acid glutaraldehyde compound disinfectant (under different dilution ratio and action time against PPV).It turned out that, PPV was exterminated under 8~128 times dilution,5 min or 2048 times dilution,30min. It demonstrated that the MBC(Minimal bactericidal concentration) of Peracetic acid glutaraldehyde compound disinfectant against PPV was 0.049%, with the shortest time sterilization 5 min.(4)By methods of HPLC and chemical titration to determined the content of the active ingredients of Peracetic acid glutaraldehyde compound disinfectant. It turned out that no significant difference (P>0.05) in the contents of peracetic acid, hydrogen peroxide and glutaraldehyde. Using HPLC to determined the chromatographic peak of peracetic acid,hydrogen peroxide and glutaraldehyde in peracetic acid and glutaraldehyde compound disinfectant, it turned out that, The degrees of separation were respectively 3.00% 3.12% and 2.50%, Repeated measurements the peak areas of each active ingredient of the disinfectant in same concentrations, the relative standard deviation (RSD) was less than 2.0%.By the add sample recovery test showed that the recovery rate was between 98.95% and 100.39%,RSD was less than 2.0%. When The concentrations of solution A and solution B in peracetic acid glutaraldehyde compound disinfectant were within the range of 0.03%~1.00% and the scope of 0.11%~0.20%, it showed a good linear relationship with each active ingredient peak areas. Peracetic acid, hydrogen peroxide and glutaraldehyde’s minimum detectable quantity in the disinfectant were 3.4ng,2.4ng and 1.0μg respectively.It showed that, by HPLC measured the content of the active components of peracetic acid glutaraldehyde compound disinfectant had a large degree of separation, high specificity,good reproducibility, high recovery rate, wide application range and can determined content of trace components.(5) Using accelerated storage test of 54℃.Determined the decline rate of the content of active components of peracetic acid glutaraldehyde compound disinfectant, which was accelerate saved 14d at 54 ℃.It turned out that,after accelerate saved 14d at 54℃ and light-free, the decline rate of content of peracetic acid,hydrogen peroxide and glutaraldehyde in peracetic acid glutaraldehyde compound disinfectant were all less than 10%.It showed that peracetic acid glutaraldehyde compound disinfectant can be saved in condition of room temperature and light-free for 1 year.
