| The purpose of this study was to determine the preparation technology of Peracetic acid glutaraldehyde compound disinfectant(hereinafter referred to as modified compound disinfectant)and the determination method of glutaric acid content,the anti-interference ability of the modified compound disinfectant to temperature and organic matter,and its bactericidal effect on Mycobacterium smegmatis.According to the methods of "Preparation of Standard Titration Solution of Chemical Reagents"(GB /T 601-2016)and "Disinfection Technical Specification",in this subject,the modified compound disinfectant prepared by different methods was compared the content of active ingredients and studied their bactericidal effects.we determined the preparation technology of compound disinfectant,the killing effect of modified compound disinfectant on Mycobacterium smegmatis,the anti-interference ability of modified compound disinfectant to temperature and organic matter were determined,and the killing effect and its influencing factors were verified;To established and verify a method of the determination of glutaric acid in compound disinfectant by HPLC.The test results showed that:(1)In peracetic acid,glutaraldehyde and hydrogen peroxide as the main raw material,hydrogen peroxide,glutaraldehyde and acetic acid as main raw materials were prepared and compound A compound disinfectant formulation B after 48 h,active ingredients peracetic acid and hydrogen content difference peroxide.That was not significant(P>0.05).The minimum effective bactericidal concentration of two kinds of compound disinfectants with different preparation procedures to Escherichia coli was 512 times diluted(0.195%),the shortest effective bactericidal action time was 5min,and the bactericidal rate was higher than 99.9% when they act on the lowest effective bactericidal concentration for 5min,all of which can achieve the bactericidal effect.The bactericidal rate was 100% when they act on 7.5min,all of which could achieve the bactericidal effect.The results showed that qualified peracetic acid glutaraldehyde compound disinfectant could be prepared by both preparation procedures.(2)Using GC-MS to determine the change product of glutaraldehyde in modified peracetic acid glutaraldehyde compound disinfectant,it was found that glutaraldehyde was oxidized to glutaric acid;The chromatographic peak resolutions of glutaric acid chromatographic peaks in compound disinfectants measured by HPLC were: 2.17 and 14.65;the relative standard deviations(RSD)of peak areas of glutaric acid chromatographic peaks in disinfectants of the same concentration were repeated 6 times and were<2.0%;the average recovery rates for the three groups were 101.3233%,100.3426% and 100.6336%,all between 98.0%-102.0%,and the recovery rates between the groups and the nine RSD were all<2.00%;the peak area of the chromatographic peak of the glutaric acid in the concentration range of 0.067 mg /m L-0.8 mg / m L showed a good linear relationship.The detection limit of glutaric acid was 19.6094 ng,and the limit of quantification was 40.6458 ng.The test showed that the method for determining the content of glutaric acid in the compound disinfectant by the high performance liquid chromatography is reliable.(3)The minimum effective bactericidal concentration of the compound disinfectant against Mycobacterium smegmatis was 512 times dilution(0.195%),and the shortest effective germicidal time corresponding to the minimum effective bactericidal concentration was 30 min.The bactericidal efficiency of the modified compound disinfectant with the minimum bactericidal concentration 512 times dilution(0.195%)for 30 min and the bactericidal rate of 100% could be achieved.(4)The modified compound disinfectant had the action of 5 minutes at the temperature of 0℃-40℃,the sterilization rate of the Escherichia coli could reach 100 percent,the sterilization rate of the hydrogen peroxide,the peracetic acid and the glutaraldehyde at the temperature of 30℃-0℃ was reduced with the temperature,and the sterilization rate was reduced with the temperature when the compound disinfectant was at the temperature of 30℃-40℃;Under the influence of 10%-50% of organic matter,the modified compound disinfectant had no effect on the killing of Escherichia coli,and the sterilization rate of 5 min could reached 100%,but with the increase of the organic matter concentration,the sterilization rate of the peracetic acid was reduced,and the sterilization effect of the glutaraldehyde did not have a significant effect.It illustrate that temperature and organic matter would not significantly affect the disinfection effect of the modified compound disinfectant. |
