Anthocyanins, which are one subclass of the polyphenols and widely distributed in plants,are synthesized by the phenylpropanoid and flavonoid pathways,not only helped coloration of plants, also can protect plants against ultraviolet radiation and a variety of pathogenic microorganisms;Moreover, the content and composition proportion of Anthocyanins play a important role in determining the quality of berry and grape wine. Anthocyanidin synthase(ANS), located at the end of biosynthesis pathway of anthocyanin, catalyzes the colorless leucoanthocyanidin to the colored anthocyanidins.In this study, with 7-years old (Vitis vinifera L. Cabernet Sauvignon) grape berries as materials, at the distinctive development stages were treated with UV-C irradiation, regarding ANS as a pointcut,to elucidate the effects of the UV-C irradiation on biosynthesis and accumulation of anthocyanins and the response mechanism of ANS on UV-C irradiation during grape berry development.The main results were as follows:1.The pattern of ANS activity during grape berry development was not changed,but the ANS activity was up-regulated significantly with UV-C irradiation 5min, while the ANS activity was down-regulated with UV-C irradiation 10min.2.The results of Western-blot showed that the protein expression level of ANS increased with UV-C irradiation 5min,while the protein content of ANS significantly reduced with UV-C irradiation 10min.3.The results of Real-time PCR showed that the expression level of VvANS was up-regulated by UV-C irradiation 5min, while the transcriptional expression of VvANS was up-regulated in young fruits and significantly inhibited in other period with UV-C irradiation 10min.4.The results of HPLC showed that the content of three anthocyanin were increased during grape berry development, the content of three anthocyanin of grape berries were significantly inhibited by UV-C irradiation, and the inhibiting effect of UV-C irradiation 10min was significantly greater than UV-C irradiation 5min.5.The correlation analysis showed that there was significantly positively related between ANS activity and ANS protein content under UV-C irradiation 5min.ANS protein content was significantly positively correlated with transcriptional expression of VvANS under CK and UV-C irradiation 10min.The transcriptional expression of VvANS was also significantly positively correlated with the total content of three kinds of anthocyanins under UV-C irradiation 10min.
