Aegilops biuncialis(2n = 4x = 28,UbUbMbMb) is a wild tetraploid line of Aegilops genus Aegilops umbellulata species. As a third gene resource of wheat, Ae. biuncialis carry many outstanding agronomical features, for example, drought tolerance, salt tolerance, insect and pest resistance, disease resistance, in addition to, it also has special high molecular weight glutenin subunits(HMW-GSs). Therefore, It can be used as an important and potential source for wheat improvement. Until now, only Molnár-Láng M and his team have carried out some work of Ae. biuncialis in recent years. So we need to further transfer the useful genes from Ae. biuncialis into wheat via screening its addition lines, substitution lines, or translocation lines.In the study, Ae. biuncialis was crossed to wheat line CN19 to produce F1 hybrid plants. and then the hybrid plants were treated with colchicine, we obtained wheat- Ae. Biuncialis amphiploid(AABBDDUUMM, 2n =10x =70). After preliminary identification of the descendants of the hybridization between CN19 and wheat- Ae. biuncialis amphiploid, we obtain a genetically stable line 12-5-2, which contains special high molecular weight glutenin subunits from Ae. biuncialis.Chromosome numbers in root-tip cells of 12-5-2 were 2n=44, and it was immune to both stripe rust and powdery mildew. It was confirmed that 12-5-2 was a 1Ub disomic addition line(ADL1Ub) by fluorescence in situ hybridization and C-banding. seed protein SDS-PAGE analysis showed that 12-5-2 not only owned all high molecular weight glutenin subunits(7 + 8 and 2 + 12) from its parent CN19, but also had 2 new subunits, which were named Ux and Uy. 22 PLUG primers from homologous group one and 26 PLUG primers that were designed were used to amplify CS, Ae. biuncialis, CN19, 12-5-2, we obtained 5 pairs 1Ub-specific PLUG primers, TNAC1021, TNAC1041, TNAC1071, TNAC1-01, and TNAC1-04. What is more, we determined the flour quality parameters of 12-5-2 and its parent CN19, it showed that the protein content, wet gluten content, grain hardness and Zeleny sedimentation value of 12-5-2 were apparently higher than those of its parent CN19.In summary, we use the methods of biochemistry, morphology cytology, and molecular biology to identity the BC3F2 descendants of hybridization 12-5-2. the new ADL1 Ub 12-5-2 could be a third gene resource for wheat improvement, in particular, it has a great advantage in improving the quality of wheat end-product and resistance to disease. It can be inferred that 12-5-2 will be high value for wheat breeding, good application of Ae. biuncialis gene pool and developing chromosome engineering breeding.
