Pseudorabies(Pseudorabies Disease)is also known as Aujeszky,disease(Aujeszky ’s, AD), is composed of pseudorabies virus(Pseudorabies, Viurs, PRV) caused by including a variety of domestic and wild animal with fever, itching(pig except) and encephalomyelitis as the main features of an acute, highly contact infectious disease, of which the most common infection in pigs. Pregnant sows mainly for abortion, stillbirth,reproductive mummified piglets when infection occurs, the mortality rate of up to 100%,mainly for increased body temperature, breathing difficulties, the nerve symptom,salivation, growth arrest, weight loss and other symptoms, boars after infection will lead to the decline in the quality of semen and even loss of seed value, and adult pig relatively mild symptoms of infection. At present, swine pseudorabies has spread throughout the world, China has more than 20 provinces and cities have the disease, and has spread and rising trend, therefore, the disease has been the world organization for animal health(OIE)classified as B class of animal epidemics, our country will also list it as two kinds of animal disease. The disease in Guangdong Province universal existence has seriously affected the healthy development of pig industry. In order to understand the epidemic situation of pseudorabies in pigs in Guangdong Province, to facilitate the prevention and control work, this thesis has carried on the investigation and research from the following aspects:1. Epidemiological study on Guangdong Province of Swine PseudorabiesThe prevalence of PR in serum of Guangdong Province during 2014 were investigated and analyzed. Look from the geographical distribution, the Pearl River Delta region of the RP prevalence rate is the highest(30.41%) followed by the western Guangdong area(21.14%), North Guangdong(11.31.%), East Guangdong(9.69%) PR prevalence is relatively low. From the time distribution, the higher the positive rate of PR months are March, August and November, the time distribution is also consistent with the results of g E-ELISA detection, and more concentrated in the winter season.2. Isolation and identification of Guangdong Province and sequence analysis of porcine pseudorabies virus strainEight PRV isolates g E, TK gene and the domestic and foreign strains of GuangdongProvince to the separation of nucleotide and amino acid homology analysis showed that the strains g E, TK, gene and 15 strains of domestic and abroad published strains of nucleotide sequence homology respectively 97.3%~99.9%, 98.7%~ 99.9%, amino acid homology respectively 94.6%~99.8%, 98.1%~99.3%; some sites have been mutated are g E, TK gene. Phylogenetic analysis showed that the strain isolated in China in recent years, with the representative isolates isolated from different provinces had a close relationship with far abroad, separation of representative strain relationship. The results of this study indicate that porcine pseudorabies virus in Guangdong area parts of isolates have been mutated, for scale pig farm in the next step of purification and selection of Swine Pseudorabies Vaccine to provide reference.3. Study on Guangdong Province pseudorabies detoxification and g E, g B antibody level of relationshipThrough the experiments of surveillance of swine pseudorabies positive field and negative field in different time sections of different swine PR virus and g E and g B antibody level, analysis the relationship of pigs in PR detoxification and antibody levels,according to the monitoring results to develop a comprehensive prevention and control measures for better containment of pig pseudorabies prevalence.
