| Based on experiments in potted, Studied the effect of different exogenous substance’s repair on Soil Ecological and different Physoilogical Indices and the main of chemical components of tobacco(protective enzyme, content of reactive oxygen species, activity of IAAOase and ATPase en ta). to explore the repairing effect of different exogenous substance on soil and tobacco under quinclorac stress.1. Repair of different concentrations of CaCl2 on the physoilogical property in leaves of Flue-cured tobacco under quinclorac stressCompared to normal leaves of Flue-cured tobacco, in leaves of Flue-cured tobacco under quinclorac stress, activity of protective enzyme, clearance rate of O-2.,.OH and rate of antioxidant declined,content of MDA and Pro increased. Spraying different concentrations of CaCl2 on leaves under quinclorac stress, could recovered activity of protective enzyme, decreased content of ROS,increased the activity of clearing ROS,reduced MDA accumulating and decreased membranous oxidation, leaped content of Pro and increased stress resistance of tobacco leaves. the suitable concentration for repair effect was 10 mmol·L-1.Compared to normal leaves, photosynthesis of suffer control leaves was inhibited, and the content of photosynthetic pigments decreased all, Fv/Fmã€Î¦PSIIã€d P decreased too, all reached significant level, NPQ increased significantly. After spraying different concentrations of CaCl2 on leaves under quinclorac stress, the content of photosynthetic pigments, Fv/Fmã€Î¦PSIIã€d P all increased,NPQ decreased,the best concentration for repair effect was 10 mmol·L-1, reached significant level.Under quinclorac stress, the content of soluble protein and activity of ATPase decreased in leaves, but activity of Cx and IAAOase increase. Spraying spraying different concentrations of CaCl2, which could increased the content of soluble protein and activity of ATPase and reduced activity of Cx and IAAOase. The repair effect as follow: 10 mmol·L-1 CaCl2> 5 mmol·L-1 CaCl2>15 mmol·L-1 CaCl2.2.Repair of different concentrations of CaCl2 on electrophoretograms in leaves of flue-cured tobacco under quinclorac stressAnalyses the SDS-PAGE of soluble proteins’ pattern, some disappeared under quinclorac stress, the protein bands of 50 k Da and 56 k Da degraded in 5d.these proteins which disappeared could recovered and some protein bands could express in advance after spraying different concentrations of CaCl2. what’s more new proteins band could express. The new protein of 120 k Da expressed by spraying5mmol?L-1 ã€10mmol?L-1 CaCl2 in 15 d later. SOD isozyme pattern showed that SOD isozymes were inhibited and some bands disappeared in leaves of tobacco under quinclorac stress. Activity of SOD-1 and SOD-2 decreased in fifth day, spraying CaCl2 could make the SOD-1 and SOD-2 enzyme recovered. Analysis of pattern of POD isozyme, activity of POD increased in leaves of tobacco under quinclorac stress, which was signally different from normal leaves. Spraying exogenous CaCl2 could repair activity of POD preferably, what’s more, made some bands express in advance. In the 15 d, POD-1disappeared, activity of POD-1 decreased in normal leaves, POD-1 expressed and POD-5 disappeared in leaves under quinclorac stress, each isozyme bands expressed and activity of isozyme was high after spraying 10mmol·L-1 CaCl2,15mmol·L-1 CaCl2.Compared to other concentrations, 10mmol·L-1 was the best concentrations for repair of soluble proteins, SOD and POD electrophoretograms.3. After spraying exogenous CaCl2, the content of total alkali and chloridion decreased, total sugar, reducing sugar and potassium significantly increased, 10mmol·L-1 CaCl2 improving the quality was better.4. Repair of different exogenous substance on physical and ecological in soil under quinclorac stressSoil pH had a little change when soil was polluted by quinclorac, adding different exogenous substances in polluted soil, p H of all treats changed. Polluted soil had no significantly difference with normal soil. Adding different exogenous substances in polluted soil,exogenous substance 1,2and 3 had no significant change, but exogenous 4 and 5 changed significantly, microbial community such as bacteria, fungi and actinomyces decreased in the soil which was polluted by quinclorac.Adding exogenous substance in quinclorac polluted soil, microbial community gradually recovered, exogenous substance 4 reached the best repair effect in all. The activity of CAT, Inv, Urase and Polyphenol oxidase decreased, protease activity first increased then decreased. Adding exogenous substance in polluted soil, The activity of CAT, Inv, Urase and Polyphenol oxidase increased, exogenous substance 4 and 1 had a better repair effect than other treats, protease activity first decreased then increased, exogenous substance 5,1 and 4’s repair effect were better.5. Repair of different exogenous substance on the physoilogical property in leaves of Flue-cured under quinclorac stress,When tobacco was impaired by quinclorac, leaves protective enzyme activities were inhibited, decrease the decomposing capacity of activated oxygen, rate of clearing O-2.,.OH and rate of antioxidant declined too, content of O-2. ã€.OH and H2O2 increased, finally leading to content of MDA increased. Spraying different exogenous substance could increase leaves protective enzyme activities, clear activity oxygen in tobacco, clearance rate of O-2.,.OH increased, tobacco producedO-2. ã€.OH and H2O2 slower, decreased membranous oxidation, reduced damage of leaves, made the impaired leaves grow normally. All of the exogenous substance had repair effect,exogenous 4 and 1 were better, and then were 5,2 and 3.When leaves were impaired by quinclorac, content of photosynthetic pigment of leaves decreased, Fv/Fmã€Î¦PSIIã€d P decreased too, all reached significant level, NPQ increased significantly, reducing light use efficiency of plant, losing photosynthetic energy. Compared to impaired leaves of tobacco, leaves’ photosynthetic pigment, Fv/Fmã€Î¦PSII and d P increased, NPQ decreased after spraying different exogenous substance. The better repair effects of different treats were exogenous substance 4, 1 and 5.The content of soluble protein and activity of ATPase decreased, IAAOase and Cx increased in leaves of tobacco under quinclorac stress. After spraying different exogenous substance, leaves soluble protein content and ATPase activity increased, IAAOase and Cx activities decreased. Exogenous substance 4 and 1 had better repair effect than others.The content of GSH and activity of ALS decreased, GST activity first increased then decreased,content of Pro increased in leaves of tobacco under quinclorac stress. Spraying different exogenous substance could increase content of GSH and Pro, activities of GST and ALS. In all, the better repair effect were exogenous substance4 and exogenous substance1.6.Repair of different exogenous substance on soluble poroteins electrophre-tograms of Flue-cured under quinclorac stressCompared to normal control, after tobacco was demaged by quinclorac,different parts had different protein expressing. Some protein bands of uppermost leaves under quinclorac stress disappeared, some new bands were induced at the same time. Spraying different exogenous substance to uppermost leaves under quinclorac stress, decreased protein expression, repaired protein bands, 10 d later, compared to normal leaves, uppermost leaves under quinclorac stress expressed the bands of 120 k Da and 104 k Da, but bands of 54 k Da and 25 k Da disappeared, and all repair treats recovered the bands of 54 k Da and 25 k Da, the same with normal control. When middle leaves were impaired by quinclorac,some protein bands disappear, and the protein expression was less than uppermost leaves. Spraying different exogenous substance could increased protein expression, recovered protein bands.30 d later, protein expression of middle leaves under quinclorac stress gradually decreased, and bands of 30 k Daã€45k Daã€50k Da and 58 k Da disappeared or low expressed. After spraying different exogenous substance, the disappeared band re-expressed, which was similar with normal middle leaves. Compared to normal roots, some big molecular protein bands and many low molecular protein bands disappeared in roots under quinclorac stress. Protein expression increased and disappeared bands recovered in repair treats. 10 d after exogenous treats,bands of 70 k Daã€104k Da and 120 k Da and bands between 10 k Da to 26 k Da wasn’t expressed, after spraying exogenous substance bands of 70 k Daã€104k Da and 120 k Da re-expressed. No matter uppermost leaves middle leaves or roots, the repair effect were as follow: exogenous 4, exogenous 1, exogenous 5, exogenous 3 and exogenous 2, exogenous 3 first had agood repair effect, but became worse than others at last..7. Repair of different exogenous substance on the main chemical composition in leaves of Flue-cured under quinclorac stressThe content of chloridion and total alkali increased and total sugar, reducing sugar and potassium decreased in leaves of tobacco under quinclorac stress. Spraying exogenous substance could decrease content of chloridion and total alkali, the content of total sugar, reducing sugar and potassium showed rising tendency, made the quality of tobacco tend to harmonious. In all, exogenous substance 5 and 4 were more similar with normal leaves, recovered the quality of tobacco better than other repair treats. |
