Abstract/Summary:
Wheat(aestivum Triticum)is one of the most important crops. Wheat yield is composed of three parameters, of which Grain weigh(GW) is an important factor.A set of common wheat(China Spring, CS)-wild emmer(Triticum dicoccoides, TDIC)chromosome arm substitution lines(CASLs) was introduced in this study. the GW of all CASLs were examined,And grain inCASL7 AL and CASL 6BS were sorted out as the big GW and small GW separately. Genes showing significantly differential expression pattern in CS,CASL7 AL and 6BS at 15 DPA(day post anthesis) were sorted out through transcriptional analysis. The information about the chromosome location and the expression of the candidate genes are as follows:1.GW of all CASLs and their parents CS, TDIC140 were examined, In order to analyze the QTLs responsible to grain weigh in wheat. Grain length, grain width and thousand grain weight of CASL7 AL were all significantly higher than CS, but the reverse is to CASL6 BS. Thus, chromosome arm 7AL and 6BS were all thought to contain QTLs related to GW.2.Transcriptional analysis of the grain of 15 DPA in CASL7AL、CASL6BS and CS were conducted.. 2005, 2134 and 778 genes showed differential expression level between CASL6BS_VS_ CASL7 AL, CASL6BS_VS_CS, CASL7AL_VS_CS separately. Then classification, functional annotation and enrichment analysis for all the differential expression genes were carried out.And then we screened ATP-β and AGP-S as the candidate gene of grain weight.3.ATP-β gene was cloned using homology-based cloning and this gene was located on chromosome arm 7AL. this gene was mainly expressed in leaf and pedicel, through Analysis of the expression pattern of ATP-β gene in different development time and tissues, The highest expression acted at 0DPA, while the origination of grain, the expression of ATP-β gene was decreasing before 15 DPA and then gradually rising. ATP-β gene was finally located in the cytoplasm by transient transformation.4. AGP-S was cloned using homology-based cloning and the gene was located on chromosome arm 7AL. AGP-S gene shows high homology between wheat and other crops. the expression pattern of AGP-S genes in different development time and tissues were analyse by RT-qPCR, we found that the gene was highly expressed in grain.The highest expression acted at the same day of 14 days old grain while as the development of grain, the expression of AGP-S gene was firstly rising before 14 DPA and then gradually decreasing. AGP-S was finally located in the cytoplasm and nucleus using transient transformation medidated.
