Endangered Mechanism And Protective Measures Of Ostrya Rehderiana Chun

Ostrya rehderiana Chun is a very narrow distribution range of wild plant, which only distributed in the West Tianmu Mountain foothills near the forest edge or forest, and only five wild species remain.Seeds’ germination rate is low in natural, and it’s hard to cutting and rhizogenesis, low seed differentiation. Therefore, it’s difficult to artificial reproduction. In 1999, Ostrya rehderiana was focus on the protection of national level. We observed the phonological phase and biotope of Ostrya rehderiana(2012-2013), and tested the germination rate. Dendrogram of the genetic similarity identified with ISSR and SRAP DNA markers among samples from wild species(Original Trees) in west of Tianmu Mountain, and introductions in Hangzhou Arboretum, provincial forestry academy of sciences,Mountain Wuchao in Hangzhou, Mountain Wuchao in Qingyuan, and Zhoushan. We are trying to offer a better way to protect the relic species. Here come the results:1. Ostrya rehderiana Chun usually Live in the permeable light acid soil with evergreen broad-leaved or conifer species. According to the local environment, the habitat belongs to a typical subtropical climate, in which annual rainfall of 1953 mm. The soil is yellow soil.2. Seeds germination test of Ostrya rehderiana(2013--2014) showed that using gibberellin treatment(400mg/L, 4h) after 2-month-low-temperature-store achieved a best germination rate(reached28%).3. ISSR and SRAP technique was used to amplify Ostrya rehderiana genome to study the genetic diversity and the genetic relationship within their specie. Cluster analysis showed that the genetic from Original Trees and samples from east arboretum are closed to Original Tree number 2. Samples from Zhoushan and provincial forestry academy of sciences are closed to Original Tree number 1. Now introductions in east arboretum grow up healthily, which suggests that Original Tree number 2 has a strong fertility.4. The seed to be picked in the autumn. Storing them in low temperature for two months. Steeping seed in 400 mg/L gibberell for 4 hours and sprouting in the artificial climate box. After waiting for its true leaf expansion into the nursery. On the other side, we should protect local environment, and find some other place like the Tianmu Mountian.
