Meat production and meat quality not only is an important economic traits of livestock meat production, but also become an important field of research in recent years. Akirinl gene is a MSTN downstream gene and negative regulated by it. Akirinl gene participate many physiological processes include animal embryonic development, muscle satellite cell proliferation and differentiation and skeletal muscle damage repair. Therefore Akirinl gene may by participating in regulation of skeletal muscle growth and development become a new candidate gene to improve livestock meat production. Studies have shown that Akirin2 gene single nucleotide Polymorphisms (SNP) with Japanese black cattle and Korean native cattle marbling score significant correlation. As a result Akirin2 gene may by participating in meat marbling score regulation become a new candidate gene to improve livestock meat quality.This study use RT-PCR and cloning sequencing method obtain Tianfu goat Akirinl and Akirin2 genes full coding sequences. Akirinl gene sequence length 480bp, include 426bp open reading frame encoding 141 amino acids, sequence submitted to NCBI obtain login number:KF515991. Akirinl protein contains 11 phosphorylation sites, phylogenetic analysis showed that Tianfu goat Akirinl gene amino acid sequence has the high similarity with cattle and sheep Akirinl gene amino acid sequence. Akirinl gene sequence length 660bp, including 579bp open reading frame encoding 192 amino acids, sequence submitted to NCBI obtain login number:KF515992. Akirin2 amino acid sequence contains 13 phosphorylation sites, phylogenetic analysis showed that Tianfu goat Akirin2 gene amino acid sequence has the high similarity with cattle Akirin2 gene amino acid sequence.RT-qPCR detected Akirinl gene in 270 days Tianfu goat and Akirin2 gene in 360 days Tianfu goat eight tissues expression situation. The results showed that Akirinl and Akirin2 genes are expressed in eight testing tissues. Akirinl gene high expression level detected in lung and spleen, low expression level detected in liver, kidney and biceps femoris muscle; In longissimus dorsi muscle, Akirinl gene expression level first increased and then decreased with age increasing. Akirin2 gene high expression level detected in lung and spleen, low expression level detected in liver, heart, rectus abdominis, biceps femoris muscle and longissimus dorsi muscle; In longissimus dorsi muscle, Akirin2 gene expression level first increased then decreased with age increasing.Westren Bloting detected Akirinl protein in 270 days Tianfu goat and Akirin2 protein in 360 days Tianfu goat eight tissues expression situation and five ages longissimus dorsi muscle Akirinl protein expression level. Results showed Akirinl protein expression only detected in the lung and biceps femoris muscle, rectus abdominis and longissimus dorsi muscle tissues. Akirin2 protein expressed in the lung and biceps femoris muscle, rectus abdominis and longissimus dorsi muscle, no expression was detected in heart, liver, spleen and kidney tissues. In longissimus dorsi muscle, Akirinl protein expression level first increased then decreased with age increasing.
