| Maize is an important crop for food and feed raw materials in the world. The Mapping and cloning of related mutant gene in the Chlorophyll biosynthesis will benefit the understanding of photosynthesis mechanism for maize as a model plant in C4 plant.In this study, a Virescent-Yellow Leaf Mutant named yglm1 was used as an experiment material, in order to understanding the genetic characteristics of this Virescent-Yellow Leaf Mutant yglm1, and mapping the gene of Virescent-Yellow Leaf Mutant yglm1,47S×yglm1, Mo17×yglml and RP128×yglml were constructed.Based on the investigation of Virescent-Yellow Leaf of the F1 and F2 populations, we can concluded that this Virescent-Yellow Leaf Mutant can be inherited stably, and the yellow character could be maintained through the whole growth period, but the level of Virescent-Yellow will be weaken in the late stage of growth compared with early growth stage. And the yglm1 performed slender leaves, slender stem and etiolated in the whole plant.Three F2 pupulations were planted and character investigation were conducted for, the separation ratios of green plants to yellow plants in F2 for 418×yglm1 with 2679 plants and Mo17xyglm1 with 1962 plants were 3:1, and fit the 3:1 by X2 test, but the separation ratios of green plants to yellow plants in F2 for RP128×yglm1 with 1687 was close to 16:1,1270 paris of SSR primers distributed within 10 Chromosomes of maize were used to detected the polymorphism between parents 478 and yglml, as result,169 pairs primers were selected which distributed in the 10 chromosomes. There were 41 pairs in Chromosomes 1,20 pairs in Chromosomes 2,18 pairs in Chromosomes 3,20 pairs in Chromosomes 4,12 pairs the Chromosomes 5,16 pairs in Chromosomes 6,9 pairs in Chromosomes 7,18 pairs in Chromosomes 8,12 pairs in Chromosomes 9, and 3 pairs in Chromosomes 10. This results givel us the genetic diversity information for 478 and yglm1.Bulked Segregation Analysis(BSA) was used for the 169 pairs of SSR primer, yellow gene pool and green gene pool were made by 20 yellow individual plants and 20 green individual plants from F2 population of 478×yglm1.The result displayed that primer umc2337,umcl586 and umc 14923 were linked with target gene in this research.Then,500 yellow individual plants from F2 population of 478×yglm1 were used for the co-separation analysis with yellow gene. Owing to the primer umc2337, umc1586, umc1492 are located in Chromosome 9, so the Virescent-Yellow gene has been located at the short arm of Chromosome 9, between umc2337 and umc1586, with the genetic distance were 2.9cM and 3.8cM to the target gene respectively. |
