| Hanyuan pepper (Zanthoxylum bungeanum ’Hanyuan’) has been well known for its high quality of:large and fruits. In 2008, it was ranked first in the list of Top Ten Agricultural Products. In the national Grain for Gree Project, Hanyuan peper can be an optimal choice due to its commercial and ecological benefits. To strength the Hanyuan peper, this thesis studied the tissue culture of Hanyuan peper by using the stem section with bud as experiment materials. The effects of different sterilizations on explants initiation rate, rooting, and seedling and transplants. The optimal medium formulas for tissue culture are studied and the results are:(1) primary culture:the explants are sterilized by 75% ethyl alcohol and 0.1 mercury chloride (HgCl2), and then they are cultivated in the modiums which are added with 6-BA, NAA, and IBA. The results reveal that the sterilization of 75% ethyl alcohol and 0.1 HgC12 (3 mins) performs best in the explants contamination, initiation and death rates, while the optimal optimal medium formula for tissue culture is MS+6-BA 1.0 mg/L+ NAA 0.1 mg/L. The initiation rate is about 48.9% and the average bud shoot height is also higher.(2) subculture:it is divided into two stages for obtaining the the optimal modiums in which the first is for shoots proliferation and the second is for rooting. The stronger seedings after primary culture are cultivated in the modiums which are updated from the MS by adding 6-BA and NAA. In the earlier stage, the optimal medium formula for proliferation is MS+6-BA 0.5 mg/L+NAA 0.1 mg/L, and the corresponding shoots proliferation rate is increased by 2.88 times; in the later stage, the optimal medium formula for rooting is MS+6-BA 0.5 mg/L+NAA 0.3 mg/L, and the average bud height is about 3.28cm with thicker stem and greener leaves.(3) Rooting:the stronger seedings after subculture are cultivated in the modiums which are updated from the 1/2MS by adding 6-BA and NAA. The optimal medium formula for rooting is 1/2MS+IBA 2.0 mg/L. With this formula, the rooting rate is 47.8%, the average number root is 2.8, the average length is 8.8mm, and the roots are uniform and the seedlings are suitable for seedling and transplant.(4) Seedling and transplants:the optimal ground substance is 2/3 yellow soil+1/3 river sand, and the corresponding survival rate is 58.9%. |
