BNS is a new type of thermo-sensitive wheat male-sterile line, and has excellent sterility and convertibility by itself, so it is of important value for the utilization of hybrid wheat and the research of genetic resources. In order to probe into its restore mechanism and to locate related genes, in this paper, fertility characteristics of BNS were observed in the generations of hybrid combinations crossing BNS over fertile normal wheat varieties, then the possible mechanism of sterility and recovery were speculated. In the basis of this the linkage groups of restoring genes were detected in the F1 generations of crossing BNS with Chinese Spring aneuploidy, and some QTLs were located by the methods of SSR molecular markers in mapping population of F2 crossing BNS over Chinese Spring. Main results are as follows:1. When BNS be Crossed over 231 wheat varieties, and their fertility phenotypes were observed in F1 generation in sterile condition, the results showed that the distribution of self-seed setting rates bagged in F1 was continuous from 0 to 95.85%., the average self-seed setting rate is 23.66%.The percentages of higher infertility combinations which self-seed setting rate was as low as BNS, and high recovery combinations which self-seed setting rate was as high as normal vareties were 25.11% and 1.41% respectively, the rest were part infertility. On these results, it was considered that BNS sterility may be the dominant on the concept of general genetics. And its restoring genes should be non-allelic of sterile genes., It was presumed that the number of sterile gene may be one pair and the restored gene may be two pairs that had different recovery ability to the sterility. BNS and Chinese Spring hybrid F1 self-seed setting rate of 3 years showed that the Chinese Spring for BNS male sterility recovery degree of relative Chinese Spring self-seed setting rate of more than 80%, so Chinese Spring is BNS high restorer line.2. With high restorer Chinese Spring Nulli-tetrasomes crossed BNS, Three consecutive years was observed, 32 combination found that all F1 self-seed setting rate is fall, also part of the combination is decrease significantly, the combination of high infertility male parents have N1A-T1 D, N1B-T1 A, N1B-T1 D, N2B-T2 D, N7B-T7 A, N7B-T7 D and Ditelosomic lines DT-1BL, explain the Nulli-tetrasomes missing chromosomes with recovery BNS fertility restoring genes or important related genes, the linkage groups is 1A, 1B, 2B and 7B.3.Used self-seed setting rate and fertile pollen rate of phenotypic BSA pools, combined SSR molecular marker to detection restoring genes linkage groups and chain tags, detected in four related chain groups of eight for polymorphism markers, and four unrelated chain groups detected no tags. Chain of polymorphic markers to detection markers in F2 mapping groups were separated, the results showed that 5 QTL loci, it located in the three related on chromosome 1B, 2B and 1 A, but 7B chromosome not detected QTL loci. 5 QTL loci, there are three restore fertile pollen sites considered to be the main effect, the other two with self-seed setting rate and fertile pollen restoration are related.These results lay a solid foundation for the sterility and recovery mechanism of BNS, and the location of the sterile and restorer genes.
