Wheat(Triticum aestivum) is one of wide cultivated grain all over the world the second food crops. The simplification of wheat cultivars caused narrow genetic basis and lack of resistance in the source. Rye(Secale cereale) is the important exogenous gene donor for wheat improvement, contains many useful genes which can be used for improving wheat yield, quality and disease resistance. Rye/wheat tralocations is one of the best methods in wheat breeding.This research use molecular markers, in situ hybridization and morphology technical means to identify the 354 progeny of 24 wheat/rye translocation lines.The results of the study are as follows:1.Using primers PSC20 H, a total of 69 positive plants were identified in 354 offspring materials, the positive rate was 19.49%. 8 pairs of specific primers of rye 1R(IAG95、PSR596、CDO580、REMS1135、PTAG1290、MWG2077、PSR162 and MWG0912) were designed, 7 of which located on the short arm and one located on the long arm of 1R chromosome; 3 pairs of specific primers of wheat 1B(BCD1124、MWG0758 and BCD1514), one located on the short arm and 2 located on the short arm of 1B chromosome; 2 pairs of specific primers of rye 5R(MWG502 and SCM138)both located on the short arm of 5R chromosome. Results showed that segment of 1R short arm occur and 1B long arm lost in W12-35-71、W12-35-71-27、W12-36-42、W12-36-43、D12-8-10、D12-128-33、D12-128-35 which were 1BL/1RS translocation lines. The segment of 5R short arm occur in W13-21-1、W13-21-2、W13-21-3、W13-21-4、W13-21-5、W13-21-6、W13-26-8、W13-46-6 which were 5RS translocation lines.2.The 69 plants with PCR-positive bands were identified by GISH, above 15 translocation lines, including six 1BL/1RS translocation lines were homozygous and one was heterzygous; eight of 5R translocation lines were homozygous.3.The 69 plants with PCR-positive bands were identified by statistic of Agronomic Characters(15 materials for translocation lines). By comparing with the substitution materials of 1R, the substitution materials of 5R, the substitution materials of Chinese Spring, the results show that some of the offspring had lower plant height, less tillers and lower seed-setting rate; the other traits were not obvious. Some of the offspring had long spike or more spikelet, can be used as the base material of wheat breeding.
