This paper mainly studied the solid fermentation with mulberry leaf as the cult-ure medium and Ganoderma sinense as the fermentation spawn. The growin-g of the mycelium was observed and the fermentation cycle was determined ac-cording to the content of polysaccharide. The chemical compositions contents ofthe GM fungal sub-stance were determined and compared with that of Gsinense and mulberry leaf. As the mainly active principle, polysaccharide was studied specially:the extracting proc-ess was optimized by orthogonal experiment and thepurification with resin was also optimized by single factor experiment, the polysaccharide after purification was teste-d for it’s antioxidation ability, finally it was isolated with DEAE-Sephadex A-25 ch-romatography. The main research results are as follows:1. The Gsinense mycelium grew well in the mulberry leaf medium(mulberry le-af 75%, bran 23%, plaster 2%, water content 60%). Mycelium recovered quickly, grew vigorously and filled the medium in 25 days. The continuous determination of the polysaccharide content showed that it firstly drop, then increaseed, finally drop, at 25th day, it was maximum.2. The study on the main compositions showed that after Gsinense fermentation, compared with Gsinense and mulberry leaf, the fungal substance ashes content inc-reased by 3.14%, to 15.11%; total sugar content decreased by 0.55%,to 5.82%; crud-e protein content increased by 6.87%, to 27.49%; fat content de-creased by 2.26%, to 2.31%; flavone content decreased by 11.63%, to 10.45%; amino acids content in-creased by 2.60%, to 13.99%; the content of lead, mercury, arsenic and cadmium ar-e lower than the national standard.3. Under the extraction condition optimized, the polysaccharide content of the GM fungal substance is 6.36%, while that of mulberry leaf is 4.79%. In the five ki-nds of optional resin, the HPD400 is the most suitable one, it can get rid of 81.04 %pigment,89.20% protein and only 20.18% polysaccharide in 2 times and 2 hours for one time. DEAE-Sephadex A-25 chromatography was usedfor isolating and purif- ying polysaccharide from Gsinense, mulberry leaf and GMfungal substance, it was showed that each of them has three components. The GM fungal substance polysac-charide can get rid of 82.12%(·OH), and inhibit 87.94%(·O2-), better than that of Gsinense and mulberry leaf.
