It was studied in the dissertation, the cultivation of Ganoderma sinense by JUNCAO technology and the biological characteristics of Gsinense mycelium; Meanwhile, the chemical compositions of Gsinense fruit body at different developm-ent stages, different parts and different culture medium were compared and analyzed; Finally, a preliminary study of the extraction, isolation and purification of the polysaccharides was undertaken in the present dissertation.1.The optimum JUNCAO medium of Ganoderma sinense was carried out as follows by the screening test, the optimum formula of cultivation composts was Dicranopteris dichotoma:39%, Miscanthus floridulu:39%, Bran:20%, Gypsum:2%, Moisture content:63%.2.The best solid cultivation condition of Gsinense mycelium was tested by orthogonal experiment. The results are as follows:the most suitable temperature is30℃, the optimum nitrogen source was yeast, the optimum carbon source was soluble starch, the optimum pH was7.0. The suitable carbon source was corn flour and the optimum nitrogen source was bean flour for liquid cultivation of Gsinense mycelia.3.The main chemical compositions of Gsinense fruit body is constantly changing during its developing process, and the change trend of different chemical components in four major development stages is also different. The main research results are as follows:both the content of crude fat and crude protein are the lowest in the period of pileus Differentiation; The crude fat content is the highest in Maturation stage while the crude protein content is in Senescence phase after spores ejected. In general, their content changes in the whole developing process keep increasing basically. However, the polysaccharide content is descending constantly. It is highest in Budding phase, lowest in Senescence phase.4.It was determined for the the extracting process of polysaccharide of Gsinense by orthogonal experiment. The results showed that:when polysaccharide were extrac-ted by hot-water extracting method, temperature was the significant effect factor on the polysaccharide extraction rate, solid-liquid ratio and extraction time was the secondary and third. The optimum extracting conditions are temperature of100℃, time of3hours and ratio of material to water1:15.5.DEAE-Sephadex A-25chromatography was used for isolating and purifing polysaccharide of Gsinense. The results indicated that:three components of GSP1, GSP2, GSP3were isolated from water-soluble polysaccharides of Gsinense.High performance gel permeation chromatography(HPGPC) was used for purity identification of GSP2. GPC atlas displayed two peaks for GSP2-1and GSP2-2. The retention time and peak area of GSP2-1is17.463min and72.03%, for GSP2-2, is19.438min and27.97%.
