The FFRC strain common carp (Cyprinus carpio) was a new carp varieties which was obtained by the artificial cultivation in recent years.It was popular with the wast number of farmers for it,s good size, fast growth, feed conversion rate, adaptable, good patience and genetic stability qualities, it,s promotion areas continues to expand, so FFRC strain common carp becoming more and more important in the main freshwater fish. As a new breed of common carp,we need constantly to protect them and being genetic conservation and improved the new varieties’ genetic quality on the basis of the original. TRAP (Target Region Amplified Polymorphism) which is a PCR-based molecular marker was proposed by Hu, since developed it has got the wide attention due to its simple, efficient, and reliable and easy to be sequenced advantages. The present study analyses the diversity of FFRC strain common carp from the phenotype level to the molecular level, It can supply the foundation for sustainable protected such resources and genetic improvement. Meanwhile, further screening TRAP markers associated with growth aslo can laid a theoretical foundation for the development and use of molecular marker-assisted breeding in FFRC strain common carp.In this study, firstly growth traits of different growthing speed carp at different ages were measured and the data was treated by correlation analysis, multiple regression analysis and path analysis. Correlation analysis showed that in slow growing families the body height has the biggest correlation coefficient with body mass while in fast growing families it was body thickness at 80 days; but after 154 days, in different growthing speed carp each of morphological traits and body mass correlation tends consistently.Path analysis showed that the 3 morphological characters’ direct effect on the body mass reached a very significant level, in slow growing families the body length has a largest direct effect on body mass while in fast growing families the body thickness has a largest direct effect on body mass at 80 days;while at 234 days the biggest direct efficiency was body length of all the families. The line regression analysis showed that the relevant index of different families all reched 0.85 at different ages, described the body length, body thickness, body height are the main traits in affecting body weight.On the molecular level, we used TRAP molecular markers to analyz the difference growth speed families. The result show that the polymorphism bands of the eight families ranged from 9.41 to 12.00, the polymorphism percentage of the eight families ranged from 52.04% to 66.02%, the polymorphism information content (PIC) of the eight families ranged from 0.19 to 0.24, the Nei’s genetic diversity and Shannon’s indexes in the eight families ranged from 0.19 to 0.24 and from 0.29 to 0.35, respectively,85# has the highest genetic parameter values, while 74# has the lowest values. Pairwise Fst and AMOVA analysis show that part from 56# and 92# (Fst=0.0516) as well as 51# and 89#(Fst=0.0956) in moderate differentiation(0.05<Fst<0.15),the rest of the families in lower genetic differentiation(Fst<0.05), the majority of genetic variability was within family (98.3%),rather than among family (1.7%). Nei’s genetic distance showed that the genetic distance between 26# and 92# was highest while the genetic distance between 85# and 89# was the lowest. The UPGMA phylogenetic tree of the eight families was constructed based on Nei’s genetic distance.The result showed that 56# and 74# were closer and clustered together,85# and 89# clustered into another group, the remaining families have a relative far relationship.The findings showed that the eight FFRC strain common carp families existing greater genetic variability and had a high level of genetic diversity, indicating that the eight families had a large selection potential.Finally, growth trait associated with body weight and body length were preliminary analyzed using twenty polymorphic TRAP markers.We found that, in slowly growing population primer combinations IGF2a-1-Trap13,IGF2a-1-odd26 are related to body weight, body length and body height and so is with fast growing population; apart from this we aslo found that in fast growing population primer combination GHR2a-3+ Eml is related to body height.Then we verify the markers obtained in extremist populations to a random population using association analysis method, Only IGF2a-1-Trap13 is related to body weight and body length of the two different growth rate populations. In our study, a possible functional gene locus related to growth was found in FFRC strain common carp by the application of TRAP markers. This provided basis for locolization of QTL concerning improtant traits and marker-assistant selection.
