Compared with other crops, wheat is almost the most important crop on the planet. There is a large amount of genetic variation in the related species of wheat, which can be used to enrich the wheat gene pool and make its genetic resources diversification. Among such relatives, rye is one of the most important related species of wheat, which owns useful genetic resource and can be used to improve the quality of wheat, for example, disease resistance and agronomic traits.Despite it is clear in theory that to transfer the better genes in the rye into the wheat is one effective way to improve the genetic basis of wheat, problems in practice make it challenging in improvement of wheat by using rye genes, such as rapid and accurate identification of rye chromatin in the wheat. In the field of wheat engineering breeding, the technique in accurate detection of rye chromatin in wheat is important and meaningful. The study is about cytology identification of the wheat cultivar mianyang 11 and rye Kustro molecular hybrids, using wheat have been reported in microsatellite primers, wheat miniature inverted-repeat transposable elements (Mites) primers and the laboratory design wheat Mite sequence primers, to extend the PCR amplification on the additional wheat-rye monomer system, and develop the specific of rye and rye chromosome molecular markers. The results are as follows:(1) Select a set of new wheat-rye monomer additional (1R-7R), part of disomic alien additional line and telosomic additional line.(2)According to the NCBI (the national biological information technology center) of wheat MITE sequence, to design 50 of PCR primers.(3)Through amplification to identify, select rye specific primers:Oleu-FN30、 Oleu-AY44、Thal-AK63、Eos-AY494、Eos-AY81、Thal-EU01、FN564427-3、 FN564436-5 and mite 3101-23.1, Xgwm153、Xgwm232、Xgwm260 and Xgwm304.(4)Find out rye chromosome specific markers:Thal-EU82、FN564427-15and mite 3101-34.1. Among them, Tha1-EU82 is 3R specific markers. FN564427-15、 mite 3101-34.1 are 4R specific markers.(5)The markers of variant implied products are:Eos-564、Pan-DQ871、 DQ537335-7、mite 2226-1.(6) Recycling the primer Oleu-FN30 with rye Kustro specific belts to cloning sequencing,compared with blast on NCBI, find that.55bp-750bp of the sequence is similar to 1545bp-2219bp of AF245032.1,and AF245032.1 is a kind of retrotransposon.So this result prove our sequence is a part of retrotransposon.The results of our research have offered the detection of rye 3R,4R chromosome for some new markers.
