Retinol-binding protein 4, RBP-4, belongs to the lipid carrier protein family, it’s a specific carrier of retinol in blood. RBP4 transports retinol from the liver to surrounding tissues. In blood, the retinol-RBP4 complex interacts with thyroxin carrier protein, and thyroxin carrier protein can efficiently prevent the complex from being filtered away when processed to glomerular filtration. Recent studies have shown RBP-4 is linked to the induction of insulin resistance and cardiovascular inflammation. It’s possible that RBP-4 could be an inflammatory factor. According to previous studies in this laboratory, RBP-4 might be an induction of swine follicular cysts.In order to further unveil the relations between rbp4 gene and follicular cysts, in this study we collected a total amount of 79 cystic ovaries and 100 normal ovaries. We observed the polymorphism of the rbp-4 gene in sow using PCR-RELP and conducted the correlation analysis between the cystic group and the normal group. As it indicated in the results, the CC genotype detected at +249-63G>C SNP locus in rbp-4 gene had a higher risk of developing follicular cysts. Hence, the detection of the genotype of rbp-4 gene at +249-63G>C SNP locus provides a guiding light in verifying whether a sow has developed follicular cysts.In this study we have also utilized the experimental data acquired to construct a kit to detect the rbp-4 gene and tested the liability of the kit afterwards. As it showed, the rbp-4 gene detection kit can predict sow individuals with a higher risk of developing follicular cysts. The kit will play an important role in clinical diagnosis and treatment of follicular cysts, breeding and selection of replacement gilts with its high specificity and repeatability. The kit has its advantages of being convenient, fast, and sensitive and does not need other special instruments. It’s readily available for mass detection of follicular cysts in sows.
