As unsaturated fatty acid in peony seed oil is extremely rich,the research and development of oil tree peony has become the new hotspot in the research of oil plants.Oil tree peony used the cultivation mode of cortex moutan production.Germination from seeds to seedlings takes at least 6months with a low production and unstable quality.Seedling production is the most important part of peony industry.The theoretical basis is rapid germination of oil tree peony seeds in container nursery.The research in seed germination characteristics and epicotyl dormancy releasing of P.ostii ’Feng Dan’、P. rockii hybrids and P.qiui is conducted from several aspects:different container treatment,GA3 with temperature treatment,constant chilling treatment,root-tip cutting treatment and AM fungi treatment.The main research results are as follows:1.GA3 treatment advanced the first rooting time and increased the rooting rate.With the increase of GA3 concentration,rooting rate increase,but high concentration inhibits rooting.0-300 mg·L-1GA3 treatment increased the rooting rate of P.ostii ’Feng Dan’、P. rockii hybrids and P.qiui,but when the concentration is 500 mg·L- 1, the rooting rate was lower than CK.300 mg·L-1 is the best treatment.2. With the temperature rise,the first rooting time of peony reduced,the rooting rate increased,but 25℃ constant treatment inhibits rooting.15-25℃ changing temperature treatment can also break epicotyl dormancy effectively.3.Chilling treatment can break epicotyl dormancy effectively.With the chilling treatment time increase,the germination of rooted seeds increased,but long chilling treatment time is bad for production application.Under the comprehensive consideration,the best chilling treatment time of P.ostii ’Feng Dan’、P. rockii hybrids and P.qiui is 30d,40d and 50d.4.Chilling treatment is the main factor of seeds germination.Under the best chilling treatment time,root-tip cutting has little influence in improving the germination rate.Under the other chilling treatment time,root-tip cutting can improve the germination rate significantly,but is still less than the germiationg rate under the best chilling treatment time.5, Epicotyls of P.ostii ’Feng Dan’ rooted seeds germinated first if the rooted seed hypocotyl diameter at the greatest point was more than 3.0 mm or if the total for lateral roots was more than 10.6.The rooting rate and the proportion of seed whose root length was more than 5 cm in new type of forest aperture disk are significantly higher than the other two aperture disks.The germination rate of rooted seeds in no-woven-cloth container is highest.As the permeability in traditional forest aperture disk is too poor,the seed rotten rate is the highest.Considering the production cost and seedling transplanting,new type of forest aperture disk and no-woven-cloth container can be widely used in production.7.AM fungi can promote growth of peony seedlings. The promoting effection of Acaulospora mellea on fresh and dry weight and dependence of P.qiui seedlings is best.8. Acaulospora mellea is the best fungi to promote morphological index of aerial part in seedlings,such as plant height,leaf length,wide leaf blade.9. Promoting effect on root length,root surface area,root volume and projection area by Glomus mosseae treatment is the best.The effect on branch strength is similar under the treatments of three AM fungis. Acaulospora mellea is the best AM fungi to promote the germination of lateral root. The performance of Glomus etunicatum treatment is not significant on all aspects.10. AM fungi can change the mineral element content in peony. Glomus mosseae can promote the N,P,K,Ca,Zn contents in the underground part of seedlings significantlay.The performance of Acaulospora mellea on promoting N,P contents in aerial part of seedlings is the best. All AM fungis can inhibi the absorption of Fe and Cu, the inhibitory effect of Glomus etunicatum is significantly.The three AM fungis have little effcetion on Mg contents in seedlings.
