Downy mildew, caused by the oomycete Plasmopara viticola (Psedoperonospora cubensis), is one of the major threats to grapevine. Many sources of resistance to P. viticola have been described in the Vitis wild species, among which is V. amurensis Rupr. (Vitaceae) a species originating from East Asia. Using’Beibinghong’selfing individuals of’Beibinghong’ to construct the genetic map of V. amurensis Rupr. QTL mapping studies were on this map and another published genetic map. The main findings are as follows:1. The 16μL SSR-PCR reaction system was established by orthogonal experimental design. It contained l0ng of template DNA,1.0μmol·L-1 of primer,0.16 mmol·L-1 of dNTPs,1U of Taq enzyme and 1.6μL of 10 ×buffer (containing Mg2+).2. All the SSR primer pairs were used for the amplification of’Beibinghong’selfing mapping population, combine the 278 SRAP we have obtained. Finaly we obtained a linkage map by joinmap3.0 in total of 382 markers were assigned to 19 linkage groups, and the total length was 2039.1 cM with an average distance of 5.3 cM between each maker.3. Using the indoor isolated leaf inoculation method to measurethe general level of resistance of 171’Beibinghong’selfing and 149’Red world’×’Shuangyou’hybrid progenies. The results is that parent’Beibinghong’showd highly resistance, among the selfing progenies,41 were immume, the ratio in total is 24%; 46 were highly resistance, the ratio in total is 26.9%; 66were resistance, the ratio in total is 38.6%and 18 were sensitive, the ratio in total is 10.5%;’Red world’and’Shuangyou’was sensitive and highly resistance respectively, among the crossing prognies,4 showed immume, the ratio in total is 2.8%; 20 showed highly resistance, the ratio in total is 14.1%; 90 showed resistance, the ratio in total is 63.4%; 24 were sensitive, the ratio in total is 16.9%and 4 were highly sensitive, the ratio in total is 2.8%.4. Based on this, QTLs for grape downy mildew resistance were detected with software MapQTL 5.0 following Interval Mapping model. We obtained 12 QTLs in total.2 QTLs were detected on’Red Globe’genetic map which were on linkage 7and linkage 15.3 QTLs on’Shuangyou’which were on linkage 16 and linkage 17.3 QTLs on ’Beibinghong’genetic map which were on linkage 4 and linkage 16.4 QTLs on intergrate map which were on linkage 7and linkage 15.
