Xylanase is more and more widely used in feed production, but different xylanase have different enzyme properties, mechanism and application effects.This study selected bacterial xylanase and trichoderma xylanase,aimed to research its some enzymology properties and application effect in wheat-diets of yellow-feather broiler.In order to provide theoretical basis for choosing more suitable xylanase for feed production, the following two tests were carried out in this study.Experiment 1:To explore enzyme properties of different sources xylanase, this study adopts the method of DNS for comparing the changes of enzyme activity of different sources xylanase in different temperature, different PH value. The results showed that two different sources of xylanase activity along with the PH value and the temperature changes were tested parabolic shape, slightly different between bacterial xylanase and trichoderma xylanase. There were the maximum activity for bacterial xylanase within 70℃ and PH value 5.0 respectively, and enzyme activity of the enzyme were the highest within 60 ℃ and PH value 5.0 for trichoderma xylanase; Under the liquid state within 80~90℃, the all activity of two xylanases were losed. At the same temperature, enzyme activity loss of bacterial xylanase was lower than trichoderma xylanase; And processed under the moisture content of 16%, there were no different for enzyme activity loss between two kinds of xylanase within 40~90℃, relative enzyme activity were more than 80% above. Therefore, trichoderma xylanase were better suited to play a role in acid environment than bacterial xylanase, however, bacterial xylanase were more suitable to play a role in pH greater than 5.5 environment.In the same condition, heat-resistant performance of bacterial xylanase was better than trichoderma xylanase.Experiment 2:In order to investigate the effects of different sources of xylanase on growth performance of yellow-broilers and its mechanism,240 one-day similar weight health yellow broilers were randomly divided into four groups, six replications, each replication have 10 birds.Control group were fed corn-soybean meal diet,test group A were fed wheat-corn-soybean meal based diet,test group B were fed based diet added 200g/t of Trichoderma xylanase and test group C were fed based diet added 200g/t of Bacterial xylanase.The enzyme activity was 30000 IU/g and the trial period was 42 days. The results showed that:Compared with the test group A, the test group B and group C increased average daily gain of broilers,and decreased feed/weight ratio significantly (P<0.05),crude protein apparent metabolic rate respectively increased by 5.90%(P<0.05),3.54%(P>0.05), the apparent metabolizable energy and apparent metabolic rate of crude fat,crude fiber, calcium,phosphorus increased significantly (P<0.05). Sugar concentration,triglycerides content,alkaline phosphatase activity, creatine kinase levels increased significantly (P<0.05),the content of serum urea nitrogen decreased significantly (P<0.05).The spleen index, the content of serum IgA, GH increased significantly (P<0.05). The relative weight of the glandular stomach and pancreas decreased significantly(P<0.05),Jejunum villus height increased by 14.81% and 11.04% respectively (P<0.05), significantly improve the jejunum villus height/crypt depth ratio of 16.61% and 16.61%(P<0.05).Thus it can be seen that wheat-corn-soybean meal based diet added different sources xylanase achieved the same effects as corn-soybean meal based diet to some extent, and the effects of trichoderma viride xylanase on improving nutrient metabolic rate,the intestinal morphology and growth performance etc were better than bacterial xylanase.
