Wheat is the most distributed crop with the highest total output, the highest trade volume. As the situation of water shortage growing, it is significant to create drought-resistant varieties. Transgenic technology is an effective way to breed a new wheat variety, which is faster and more purposeful than traditional breeding technology. Under natural drought conditions, the observation of physiological and biochemical indexes and agronomic traits about drought resistance were conducted among the sixth generation of the new wheat trains with TaTPK gene and the drought resistance identification was determined. The results of this paper are follow:1. Through the drought resistance identification of a wheat strain 11W284-8-3-2 cultivated protein kinase gene TaTPK into kenong199, the results showed the drought resistance of the transgenic strain is significantly higher than the receptor control kenong199.(1) Under natural drought condition, the photosynthetic rate, chlorophyll content, relative water content, and water use efficiency of the transgenic stain 11W284-8-3-2 and kenong199 showed a declining trend at the whole stages of heading, anthesis and filling, but 11W284-8-3-2 kept a higher level, The transpiration rate, stomatal conductance, relative electric conductivity, malondialdehyde, content of soluble protein and proline, and the active of POD and SOD showed a trend of exaltation. 11W284-8-3-2 kept significantly lower transpiration rate and stomatal conductance, relative electric conductivity, malondialdehyde than receptor control, and the content of soluble protein and proline and the activity of SOD and POD of 11W284-8-3-2 kept significantly or extremely significantly higher level than kenong199. This clarified the transgenic lines 11W284-8-3-2 kept higher drought resistance.(2) The plant height, area of flag leaf and tiller number of 11W284-8-3-2 were significantly higher than kenong199. So were the traits of panicle. The production of 11W284-8-3-2 with 527.04 kg per mu was was more than kenong199 by 11.35% with the production of 473.33 kg per mu. This showed the transgenic lines kep thigher drought resistance in agronomic traits.2. Through the drought resistance identification of the wheat strains 12SR494-1-2、12SR500-7-4 were conducted by integrating linear gene TaTPK into jimai 22. The results showed the drought resistance of the transgenic strains were significantly higher than the receptor control jimai22.(1) Under natural drought stress, the photosynthetic rate, chlorophyll content, relative water content, water use efficiency stayed reducing at the whole stages of heading, anthesis and filling, but 12SR494-1-2, 12SR500-7-4 kept higher level than jimai22. The transpiration rate, stomatal conductance, relative electric conductivity, malondialdehyde, content of soluble protein, proline, and the active of POD and SOD showed a trend of exaltation. The transpiration rate, stomatal conductance, malondiadehyde content, relative electric conductivity of 12SR494-1-2, 12SR500-7-4 kept a lower level, and the soluble protein content, proline content and the activity of SOD, POD were significantly or extremely significantly higher than control. This showed the transgenic lines could effectually kept higher drought resistance.(2) The area of flag leaves, and the length of kernel of 12SR494-1-2, 12SR500-7-4 were significantly higher than jimai22. So were panicle traits such as kernels per spike and kernels per plant. The production per mu of 12SR494-1-2, 12SR500-7-4 was 688.15 kg, 700.18 kg and higher than jimai22 by 10.26% and12.29%, with the production 624.07 kg per mu. This showed the transgenic lines kep thigher drought resistance.
