Quercus variabilis, a source of industrial cork, is one of the most valuable Quercus species native to China, which could produce the timber, fuelwood, edible fungus, Tianma,acorns, oak bowl(tannin). It is propagated mainly by seedling transplantation which results in extensive genotypic and phenotypic variations. Furthermore, seed viability can be as low as mature acorns and are prone to be attacked by several insects and diseases before collection and during storage, as well as seeds being recalcitrant for storage. A potential solution to these problems would be the development of methods for vegetative propagation of this species. However, trees can be shown to have excellent genotype when it has become a big tree with a mature character, which is poor rooting and weak growth. Thus it brings great difficulties to extended use excellent genotypes with vegetative propagation. In the present study, we tried to define some possible differences in the protein composition of juvenile and adult Q. variabilis plants under well defined genetic and developmental stages. It will provides a theoretical basis for deep understanding phase change mechanism and rejuvenation in Q. variabilis. The results are as follows:1.The experiment compared trichloroacetic acid(TCA) method / acetone(acetone) 、phenol extraction method、acetone precipitation method in total protein content、SDS-PAGE、Two-dimensional electrophoresis. The results showed that the phenol extraction method extract the maximum total protein; the protein obtained the largest number of protein banding in unidirectional SDS-PAGE; the protein separated by two-dimensional electrophoresis, and2 D map resolution is good; the spots is clear, which can effectively resolve Quercus proteome.Thus, the present study established a two-dimensional electrophoresis system.2.The experiment have find the best extraction time of leaves protein in Quercus. By comparing protein content and the pattern of SDS-PAGE in different seasons. The results showed that: The protein content at the beginning of October is the highest and the components is most than other months.3.The experiment determined the genotype does not affect the protein content in Quercus.Comparing protein content and the pattern of SDS-PAGE in nine different genotypes in three ages. The results showed that: in different genotypes of the same age, the protein content have no significant difference,but different age class have significant difference.4.The experiment uses different parts of different age class for SDS, by electrophoresis can find the protein in the base of the same age class significantly more than the crown. The the molecular weight of 14.4 k D、28.4 k D、53.4 k D、66.0 k D protein only appear in the base,The the molecular weight of 15.8 k D、20.1 k D、27.0 k D、29.5 k D、30.0 k D、42.2 k D、45.0k D、50.6 k D、61.8 k D、104.8 k D protein only appear in the crown. The protein content in different age classes of the base was no significant difference. The crown in different age classes have quite different protein content. Protein in lower age class was significantly higher than younger age classes. Base protein bands significantly thicker than the crown, dark in color.15.8 k D、20.1 k D、27.0 k D、29.5 k D、30.0 k D、42.2 k D、45.0 k D、50.6 k D、61.8 k D、104.8 k D belong to albumin, 14.4 k D、28.4 k D、66.0 k D belong to globulin, 53.4 k D belong to gliadin.5.The experiment uses the two-dimensional electrophoresis analysis the material of base and crown in the first age class, By electrophoresis patterns it can be found that protein spots were significantly higher inbase than in crown.
