The technology of Plant Regeneration from the Zucchini in vitro female nucl eus is an important way to obtain the germplasm resources. Similar studies have been carried out at home and abroad, but there has been no successful application. The m ain problem is that the technical repeatability is poor, the embryo rate is low, vitrifica tion phenomenon is obvious and so on. The aim of the research was to guarantee nor mal production of the embryoid, minimize the impact of vitrification on plantlet regen eration from megaspore and enhance the survival rate of test-tube plantlets. This exper iment has choosed two varieties of zucchini were used as material, Compared with th e unfertilized ovary of unfertilized ovules embryo effect; Screened of the best culture medium for four kinds of solid medium; Researched the effect of training environment,exogenous hormone concentration on vitrification phenomenon influence on culture bo dy; Studyed the doubling effect of two kinds of liquid and the method of domesticati on and transplanting of regenerated plants,etc. These results have important reference v alue on optimize zucchini unfertilized ovary induce regeneration plant technology. The main results were as follows:1. In the same medium zucchini unfertilized ovary the effect of the embryo was better than that of the unfertilized ovules. Unfertilized ovules in addition to the individual color turn green, the rest is not any change; and unfertilized ovary has different level volume enlargement, the color turned green.2. Under the condition of high 35 ℃heat shock 6 d effect best, ovule volume big, dark green color; 5 d volume is too big, but the color is light green; 4 d compared with small volume, color is yellow.3. In solid medium:B5+40g/LSuc+8g/LAgar+0.5mg/LNAA+0.05mg/LTDZ+30mg/LAgNO3 with highest rate of embryo.4. When the environment is 25℃,the embryo rate is 60%,vitrification rate is 20%,produce vitrification phenomenon is the weakest,the comparison is the best cultivation of environment temperature.5. Environmental humidity was 95%, of vitrification phenomenon is more serious; 85% humidity test tube seedlings healthy growth, produce the vitrification rate is low, is the best training environment humidity.6. Medium added GA3 concentration of 0.5 mg/L to induce rooting the highest survival rate, at 46.67%, and vitrification phenomenon is the weakest.7. Zucchini in vitro female nucleus induction training in 85% humidity, illumination 12 ~ 14 h/d, the light intensity of 1200 ~ 1500 Lx, 25 ℃, rooted in the culture medium when adding GA3 concentration is 0.5 mg/L, can effectively reduce the incidence of vitrification phenomenon, promote the health of the tube seedlings to grow.
