| Carpinus cordata belongs to the Betulaceae Carpinus plants, which is a kind of excellent wild ornamental plant resources. The crown shape is beautiful, the ornamental effect is good, without serious diseases and insect pests. C. cordata is suitable for the city’s ecological environment, can be used as street trees and garden tree species, especially suitable for high hedgerow trees. Used in urban greening, C. cordata can increase the species of garden trees, and rich urban landscape.With a method of comparison and contrast between different concentration of GA3, different storage methods, this research studies the effects that different ways of treating seed had on the germination rate of C. cordata seeds. The seeds of C. cordata were tested and compared with the seed germination of the seeds treated by direct sowing and sowing after hormone plus storage. The ordinary thin branches and the thick branches germinating in second year after pruning branches in great intensity in wintter, treated by different concentrations of IBA and NAA, to explore the effects of different treatments on the survival of cuttings of C. cordata. Leaves, buds, roots and seeds of C. cordata were selected as explants, testing the method of surface disinfection of explants, to explore the best disinfection time and ways of leaves, buds, roots and seeds. To explore the culture medium formula of callus production for leaves and roots, and to explore the feasibility of the buds become propagation material. To explore the formula of the culture medium for inducting the seeds differentiate directly into callus, inducting the leaves of seedling without virus into callus, and for inducting the seedling grow roots. Finally, the following conclusions are drawn.â‘´Both GA3 and wet sand storage have a good effect on the germination rate. Different doses of GA3 and different storage ways have different effect on germination rate of C. cordata seeds; Using GA3 before storing the seeds has a better result than the other way round. Selecting the mature and full seeds, first contact them with 1000 mg/L GA3 for 12 h, then store them in the sand pit outdoor, with this particular way, the germination rate would be the highest.⑵For the C.cordata seeds, the better the seed maturity, the more weight, the higher the germination rate. At the beginning of October, the maturity of C.cordata seeds is the best, and the germination rate is the highest.In the end of August, the maturity of C. cordata seeds is not enough, and the germination rate is low.â‘¶After germination treatment, seeds of C.cordata of sowing after outgrowing the hypocotyl have a high mortality rate, the key point of increasing the seedling rate of C. cordata seeds is controlling the degree of germination of seeds.â‘·The method of soaking the branches for 5 minã€15 min and 30 min respectively with the solution of orthogonal combination of 300 mg/kgã€600 mg/kgã€900 mg/kg concentration of IBA and 600 mg/kgã€900 mg/kgã€1200 mg/kg concentration of NAA, had no obvious effect on rooting the branches of C.cordata.C. cordata is difficulty of cutting propagation.After pruning branches in great intensity in wintter, survival time of the germination of thick branches in second year is longer than the ordinary thin branches.⑸The best way to disinfect the leavesã€buds and roots of C. cordata is: after disinfecting for 1 min by 75% alcohol, disinfect for 9 min30sã€6 minã€7 min respectively by HgCl2.⑹The leavesã€buds and roots of C.cordata are difficult to carry out tissue culture, explants are prone to browning, in contrast, the seeds of C.cordata are the best explants.⑺GA3ã€6-BA and IBA are conducive to developing the seeds of C.cordata to seedling.â‘»Seed reproduction is the most economic and convenient way in C. cordata breeding methods. Selecting the mature and full seeds, first contact them with 1000 mg/L GA3 for 12 h, then store them in the sand pit outdoor, take out the seeds and seeding when the seed coats are dehiscent, with this particular way, the seedling rate would be improved. |
