Plants of Crassulaceae as an excellent ground covers have the ornamental value、 good resistanee and patience. Sedum selskianum’Spirit’、Sedum spurium’Coccineum’、 Orostachys malacophyllus were selected to study their efficient propagation methods and develop an efficient procedure of plant regeneration of Crassulaceae plants. Having significan ce to ensure the sustainability of development and utilization of plants of Crassulaceae. Test were investigated from seed germination、cutting propagation and tissue culture separately. The study results were showed as follows:1.Different temperatures have different effects on the seed germination of different seeds of Crassulaceae. The temperature which too high or too low are detrimental to the seed germination. The optimum temperatures for the seeds germination of Orostachys malacophyllus and Sedum spurium’Coccineum’were25℃.The optimum temperatures for the seeds germination of Sedum selskianum’Spirit’was25℃-30℃.2.The stem segment propagation technique of Sedum spurium’Coccineum’was also explored in this study by testing various factors including cutting media、different kinds of plant growth regulators、plant growth regulators in different concentrations and different temperatures. The results showed that the optimum environmental condition for cuttage propagation was soaking in150mg-L-1NAA before cutting, In the media of1:1perlite and vermieulite, at20℃-25℃.3. Leaves, stems and and leaves with axillary buds, stems with axillary buds of Orostachys malacophyllus were selected to study the effect of explants, different plant growth regulators and different environmental condition on tissue culture of Orostachys malacophyllus. The results showed that treated with75%aleohol for30s and followed by0.1%HgCL2for5min was the optimum explants disinfection of leaves and leaves with axillary buds of Orostachys malacophyllus. Treated with75%aleohol for30s and followed by0.1%HgCL2for7min was the optimum explants disinfection of stems and stems with axillary buds of Orostachys malacophyllus. No significant variation was found on callus induction from leaf of Orostachys malacophyllus under different basic media and different pH. The optimum conditions for callus induction were under light treatments. The medium of callus induction for leaf:MS+6-BA3.0mg·L-1+NAA0.1mg·L-1was suitable, in which the callus induction rate was83.33%.The medium of callus induction for stem:MS+6-BA3.0mg·L-1+NAA0.3mg·L-1was suitable. Initial medium of the leaves with buds and stems with axillary buds:MS+6-BA1.0mg·L-1+NAA0.4mg-L. The optional culture medium combination for calluses proliferation was:MS+6-BA0.5mg·L-1+NAA0.1mg·L-1+2,4-D0.01mg·L-1,the proliferation rate was2.69. The optional culture medium combination for buds proliferation was:MS+6-BA1.50mg·L-1+NAA0.4mg·L-1,proliferation index was5.35. The optimum medium for rooting was1/2MS+NAA0.3mg·L-1,the best rooting rate could reach100%. Forging seeding transplanting:The transplant medium in perlite,vermieulite,and garden mould by1:1:1was the optimal for Orostachys malacophyllus,which the rate of survival was76.67%.
