| Jute,which also called green hemp, belongs to Tiliaceae. Jute annual herbaceous bast fiber crops. The main cultivars are:Corchorus capsularis L. and Corchorus olitorius L. Since jute fiber having some advantages of high yield, soft, good moisture absorption,and so on, so which was widely used in commercial, industrial and construction areas.So the study of jute tissue culture techniques are increasingly important, materials come from tissue culture, the process provide a reference for the comprehensive utilization of jute. In this experiment, young stems were using as explants in jute tissue culture, induction of embryonic callus, plant regeneration system and the establishment of embryogenic cell suspension system. We obtained the following results from this experiment:1 Tissue Culture and Regeneration System(1) To decide callus induction medium of jute, Corchorus capsularis L. and Corchorus olitorius L.were used as materials, young stems were explants, with the Uniform Design to inducte callus. The results showed that the callus induction of jute is 179> MeiFeng fourth> longxi long fruit> GuangBaAi.And the callus induction medium of Jute 179 is MS+0.53 mg/L 2,4-D+2.0 mg/L 6-BA, which of Mei Feng fourth is MS+2.0 mg/L TDZ+0.5 mg/L 2,4-D, which of longxi long fruit jute is MS+2.0 mg/L TDZ+2.0 mg/L 6-BA, which of GuangBaAi is MS+2.0mg/L TDZ+2.0 mg/L 2,4-D+2.0 mg/L 6-BA.(2) Some growth good jute callus of four genotypes were selected as explant material, adventitious buds and roots were induced with using orthogonal design, we studied the influence of genotype and hormone combinations in the process. The results showed that the regenerative capacity differences of jute callus 179> MeiFeng fourth> longxi long fruit> GuangBaAi. Jute 179 and fourth Meifeng get regenerated, longxi long fruit were induced adventitious roots, GuangBaAi only get adventitious buds. Hormone combinations on callus jute equally significant, jute 179 and Meifeng fourth were induced adventitious buds in MS+1.5 mg/L 6-BA+0.5 mg/L NAA+ 1.5 g/L Vc,and were induced adventitious roots in MS+2.0 mg/L NAA+0.5 mg/ L (1.0 mg/L) IBA, with growing well. The best medium for adventitious buds of longxi long fruit induction is:MS+1.5 mg/L 6-BA+0.5 mg/L NAA+1.5 g/L, inducing rate of 22.6%.2 embryonic cell suspension system(1) With well-grown callus of Corchorus capsularis L.,in the embryonic callus induction process to study the influence of genotype and hormone combinations. The results showed that it is easier that 179 Jute callus transition to embryonic callus than Meifeng fourth, good quality and embryonic callus induction time is short. Two varieties of jute callus induction in MS+0.1 mg/L TDZ+0.3 mg/L6-BA+0.3 mg /L NAA on the best induction rate was 54.62% and 43.88%, respectively.(2) The well-grown embryonic callus were used as materials to build suspension cell line to study the influence of culture medium, inoculum, medium volume, pH, genotype. The results showed that, jute embryogenic callus were cultivated on B5 medium, medium volume was 40mL, inoculation amount was 1.0 mL.We successfully obtain a cell suspension cell lines. Different genotypes of jute in the suspension of cell proliferation and solution pH is significantly difference. |
